Description
CFX96 REALTIME PCR W/LAPTOP MEC for qRT-PCR machine (BRAND NAME OR EQUAL) MUST BE COMPATIBLE with current system BioRad CFX96 and CFX Manager software Must be compatible with low profile (15.48mm), thin walled 0.2mL 8-tube strips with optical flat cap strips - for reaction volumes of 5-125 L Real-time data traces can be viewed during a run from the thermal cycler base Automated lid that opens, closes and provides sealing force to reaction wells Thermal gradient (differences up to 24 C front to back) allowing optimization of multiple temperatures in a single assay Peltier-driven thermocycler with max ramping speed of 4-6C/sec Thermal block range of 0-100 C Sample block accuracy of +/- 0.2-0.3 C of programmed target at 90 C with uniformity of +/-0.4-0.5 C well-to-well within 5-10seconds of arrival at 90 C Excitation and detection of at least five fluorescent dyes in one reaction Independent illumination and detection of each well and use of 5-6 LEDs and photodiodes to emit and detect (respectively). Fixed optical path over each well so there is no need to normalize based on position. Excitation with absorption spectra between the 450-684nm range; optimal detection of dye with emission spectra of 515-730nm. Capability for FRET experiments- absorption spectra in the 450-490nm range and detection with emission spectra in the 560-580nm range. System can read all 96 wells in all channels in 10-12seconds; has a fast scan mode that can read in 2-3seconds Can detect down to one copy of target sequence in human genomic DNA Reaction volumes 1-50 L; ideal of 25 L Detects less than 8-10fmol of fluorescein Must include Windows 7 laptop computer compatible with CFX96 touch qRT-PCR machine and CFX Manager software version 3.1; processor - Intel i5 or better; Hard drive- 400-500GB; RAM: 3-4GB; DVD/CD-RW drive; minimum resolution 1280x1024, preferably 1920x1080; and Adobe PDF reader. MEC for Software MEC for Software (BRAND NAME OR EQUAL) MUST BE COMPATIBLE with current system CFX96 Capable of multiplex amplification and melt curve and end-point analyses on at least 5 different fluorophores per well Can analyze multiple experiments and generate data from one plate Can add thermal cycler repeat steps can be added Can display customized run results and multiple window panes can be viewed at one time Can display multiple standard curves for more than one flour when doing multiplexed reactions Quantifies PCR by standard curve and automatically calculates reaction efficiencies at the y-intercept Can discriminate between alleles and quantifies end-point fluorescence to determine cycle (Cq) Analyzes gene expression by relative quantification ( Cq) or normalized expression ( Cq) Can assign multiple reference genes in normalized expression analysis Calculates stability and coefficient of variation for reference genes Can compare 4,000- 5,000 Cq values from different files for gene expression studies Can multiplex analyses for up to 4-5 fluorophores Exports data graphs and tables into Microsoft Excel and data can be customized before being exported Can create reports with customized information (tables, graphs, analyses) in a chosen order in PDF format.
