THIS AMENDMENT NO. 2 - IS TO EXTEND THE DATE OF ALL QUOTES BEING SUBMITTED BY (NO LATER THAN APRIL 10, 2016) ______________________________________________________________________ THIS IS A AMENDMENT TO THE FOLLOWING USDA REQUIREMENT: TO ANSWER THE FOLLOWING 4 VENDOR QUESTIONS: VENDOR QUESTION NO. 1: WHAT APPLICATION WILL THE USDA LAB/END-USER BE LOOKING TO AUTOMATE ? USDA'S ANSWER TO THE ABOVE QUESTION: USDA IS AUTOMATING THE PCR SETUP, KASP ASSAY SETUP, NGS LIBRARY CONSTRUCTION, SERIAL DILUTION SETUP, AND OTHER MORE GENERALIZED LIQUID TRANSFER TASKS - E.G. - (REPLICATION OF TEMPLATE DNA CROSS MULTIPLE REACTION PLATES). THESE APPLICATIONS WILL BE PERFORMED IN ANSI STANDARD 96, 384 AND 1536-WELL MICROTITER PLATE FORMATS. VENDOR QUESTION NO. 2: WILL USDA BE ABLE TO PROVIDE A COPY OF THE PROTOCOL THAT THEY ARE CURRENTLY PERFORMING MANUALLY OR OF THE PROTOCOLTHEY WOULD LIKE TO AUTOMATE ? USDA ANSWER TO QUESTION NO. 2: USDA'S PROTOCOLS REQUIRE THE FOLLOWING IN TERMS OF LIQUID HANDLING INSTRUMENT CAPABILITIES: A. PROVISION FOR DECK OR STAGE SPACE FOR A MINIMUM OF SIX ANSI STANDARD96, 384 AND 1536-WELL MICROTITER PLATES. B. THE ABILITY TO EMPLOY A PIPETING HEAD WITH 384 INDIVIDUAL PIPETTING CHANNELS/TIPS. C. THE ABILITY TO ASPIRATE AND DISPENSE ASSAY MASTER MIX AND/OR DNA SOLUTIONS IN VOLUMES AS LOW AS 0.5 uL AND AS HIGH AS 125 uL PER PIPETTING CHANNEL AVERAGE DISPENSING PRECISION WITH A CV<3% AT 1 uL. USDA'S ANSWER: THE INSTRUMENT BELOW WILL BE LOCATED AT THE FOLLOWING LOCATION: USDA, ARS, PLANT SCIENCE RESEARCH UNIT 4118 WILLIAMS HALL 101 DERIEUX PLACE RALEIGH, NORTH CAROLINA 27695 ____________________________________________________________________ This is a combined synopsis/solicitation for The United States Department of Agriculture, Agricultural Research Service, Plant Science Research Unit, located in Raleigh, North Carolina, requires an instrument for performing automated liquid handling. This equipment would be used to array plant DNA and reagents across assay plates in conjunction with the Plant Science Research Unit's genetic investigation of complex traits of plants. In order to determine the genetic basis of quantitative traits, DNA is isolated and marker analyses performed for thousands plant DNA samples each year. Minimum Equipment Requirements: Must be able to accurately dispense into 96, 384, and 1536-well plate formats. Must possess a minimum of six deck positions to accommodate standard footprint PCR plates. Must be able to accept a pipetting head with 384 individual pipetting channels/tips. Must be able to dispense a minimum volume of 0.5 μl and a maximum of at least 125 μl per pipetting channel/tip. Must exhibit average dispensing precision with a CV < 3% at 1μl. Must include control software with a graphical user interface (GUI) for programming automated operation. Must include an on deck tip washing station integrated with the GUI. Must include a manufacturer's warranty of at least 12 months in length. Must include a minimum 30-day site application test period to evaluate functionality.
