- This is a notice of intent, not a request for a quotation. A solicitation document will not be issued and quotations will not be requested. The Government intends to award a fixed price purchase order to Fisher Scientific, Hanover Park, IL. The Department of Transfusion (DTM) at the National Institutes of Health (NIH) Clinical Center (CC), Bethesda, MD has a requirement to procure the supplies listed below: The Cell Processing Section (CPS) is responsible for the manufacture of human cells and tissue based products utilized in phase I/II clinical protocols for transplantation, adoptive immunotherapy and gene therapy conducted in the Clinical Center at the NIH. Cell processing procedures are tailored to each of the 30 or more different patient-care clinical research protocols supported. Also, regarding Institute research support it: Develops biologics collection and manufacturing processes to support clinical trials Collects, manufacturers and distribute blood components for in vitro research use Collaboration in the design, planning execution and data analysis of clinical studies Collects and manufacturer specialized cellular products to support clinical trials Develops and evaluate assays for clinical application and biologics safety testing and cellular characterization. Primary Items sought: QuantityCatalogDescription 1SK-150-FCSCellometer Vision brightfield/fluorescence cellprofiling system, with motorized filter set switching. Comprises instrument, software, automated controller, 2 fluorescence optics modules, and a pack of 75 slides. Includes 2 licenses of FCS Express software. Presently, there is an urgent need for characterization of cellular therapeutics, including the starting material, intermediate products and final products. Especially for cellular product with high concentration of nucleated Red Blood Cells (RBCs), such as bone marrow and cord blood, measuring the concentration of white blood cells (WBC) is of critical clinical relevance; otherwise the patients may be over-dosed hence excessive cytotoxicity may be caused, or clinical expectation may not be realized due to or under-dosed cellular product. The Cellometer Vison CBA is an image cytometer for automated image capture and analysis. It uses fluorescent dye to differentiate cells types; fluorescent positive live and dead nucleated cells are easily counted and red blood cells are excluded from cell counts. There is no need to lyse red blood cells, saving time and eliminating an extra sample preparation step. In preliminary validation runs that were performed in CPS, Cellometer Vision CBA provided similar performance to flow cytometry assay; it proved to be an effective platform for counting bone marrow samples. If we cannot obtain the same system, we will have to re-run the validation on the equipment from other vendors, it will take at least a couple of months, and a lot of government funding and resources will be spent on it.
