- This is a Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to perform a potential requirement. This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition. It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract. Responses will not be considered as proposals or quotes. No award will be made as a result of this notice. The Government will NOT be responsible for any costs incurred by the respondents to this notice. This notice is strictly for research and information purposes only. Background: The mission of the National Center for Advancing Translational Sciences (NCATS) is to develop new therapeutics for treatments of rare diseases by using small molecule screening to identify lead compounds, study their mechanisms of action, and identify molecular targets. The new Stem Cell Translations Laboratory (SCTL) is part of The National Center for Advancing Translational Sciences (NCATS). Cultured pluripotent stem cells can self-renew extensively and be differentiated into all cell types of the human body. The SCTL uses a multi-pronged scientific and technological strategy to provide novel insights into the biology of pluripotent stem cells with the ultimate goal to develop novel medicines and regenerative medicine applications for diseases with unmet need. The mission of SCTL is to generate standards and differentiation protocols in order to produce large numbers of relevant human cell types (e.g. dopamine neurons lost in Parkinson's disease). Increasing the robustness and reproducibility of human stem cell protocols and implementing standard operating procedures (SOPs) are of major importance to the entire field. In addition, precise characterization of various cell type identities is intended. The SCTL will therefore investigate protein expression of stem cells on the single-cell level using the most powerful technology. This cutting-edge high-throughput single-cell proteomis is the method of choice for studying molecular signatures and cell signaling at an unprecedented resolution. The Helios enables the most comprehensive understanding of cell phenotypes and will be key equipment for the SCTL in order to accomplish its mission by generating reference datasets for the intramural and extramural scientific communities. Purpose and Objectives: The purpose of the proposed requirement is to procure a Helio's CyTOF Unit or the equivalent. The newly established Stem Cell Translation Laboratory (SCTL) will serve as a national and international reference center for stem cell research. As part of this mission, the SCTL will take advantage of cutting-edge technologies and perform protein expression studies on the single-cell level. Detailed characterization and phenotyping stem cells and their differentiated progeny at different time points will allow meaningful systems-biology understanding of fundamental cellular processes. Mapping molecular signatures in a defined cell type-specific fashion is essential to precisely capture cellular states and phenotypes. Project requirements: The required unit must meet the minimum following requirements: • Inductively coupled plasma ion source with free running RF generator and a balanced load coil interface. Inductively coupled plasma is used to atomize and ionize cells stained with metal-conjugated antibodies and metallointercalators. The atomic ions are extracted into the ion optics and time-of-flight regions where they are separated by mass and quantified • 5 stage vacuum system with 2-stage plasma-vacuum interface and turbo-pump; • Ion neutral decoupling ion deflector, low-mass cut-off quadrupole and point-to-parallel focusing lens system • Orthogonal analyzer operated at >75000 spectra/second cycle frequency • Discrete dynode ion detector • Over 50 tags may be analyzed simultaneously on a single stained sample, no re-staining or tagging is required • Detection system based on dual 8-bit signal digitizing boards • Direct sampling spray chamber for individual cell assays • Micro-concentric nebulizer • Pneumatic sample introduction system • Instrument control and data handling computer system • Instrument control and data handling software These unique attributes are required for our planned single cell proteomics research goals. Anticipated period of performance: Delivery is required within 10 weeks of receipt of the award. Capability statement /information sought. The respondents must include enough information to clearly demonstrate that they can either sell and deliver the Helio's CyTOF System or has an equivalent system that meets or exceeds the specifications stated above. The respondent must also provide their DUNS number, organization name, address, point of contact, and size and type of business (e.g., 8(a), HubZone, etc., pursuant to the applicable NAICS code and any other information that may be helpful in developing or finalizing the acquisition requirements. One (1) copy of the response is required and must be in Microsoft Word or Adobe PDF format using 11-point or 12-point font, 8-1/2" x 11" paper size, with 1" top, bottom, left and right margins, and with single or double spacing. The information submitted must be must be in and outline format that addresses each of the elements of the project requirement and in the capability statement /information sought paragraphs stated herein. A cover page and an executive summary may be included but is not required. The response is limited to ten (10) page limit. The 10-page limit does not include the cover page, executive summary, or references, if requested. The response must include the respondents' technical and administrative points of contact, including names, titles, addresses, telephone and fax numbers, and e-mail addresses. All responses to this notice must be submitted electronically to the Contract Specialist and Contracting Officer. Facsimile responses are NOT accepted. The response must be submitted to Mr. Brian Lind at lindbj@nida.nih.gov. The response must be received on or before July 28, 2016 at 9:00 AM Eastern Time. The Government reserves the right to use information provided by respondents for any purpose deemed necessary and legally appropriate. Any organization responding to this notice should ensure that its response is complete and sufficiently detailed to allow the Government to determine the organization's qualifications to perform the work. Respondents are advised that the Government is under no obligation to acknowledge receipt of the information received or provide feedback to respondents with respect to any information submitted. After a review of the responses received, a pre solicitation synopsis and solicitation may be published in Federal Business Opportunities. However, responses to this notice will not be considered adequate responses to a solicitation. Confidentiality: No proprietary, classified, confidential, or sensitive information should be included in your response. The Government reserves the right to use any non-proprietary technical information in any resultant solicitation(s)."
