- Statement for Genotyping By Sequencing (GBS) Order Sugarcane Production Research Unit USDA-ARS, Canal Point, FL 33438 A total of 400 Saccharum hybrids clones will be genotyped using Next Generation Sequencing (NGS) based GBS technique genome Wide Association Study (GWAS). One sequencing lane will not run more than 48 samples at a time. This work will include library preparation, pair end sequencing using Hiseq genome sequencer, and subsequent bioinformatics. Bioinformatics includes removal of low quality reads, alignment of reads to Sorghum bicolor reference genome and without reference genome, statistics of tag data productions, and genomic variants identification. Data needs to be delivered in the form of raw sequence data and SNPs genotypes for GWAS Analysis. Final data needs to be delivered by the 60 days from the genomic DNA accepted by the facility.
