- National Cancer Institute (NCI), Division of Cancer Epidemiology & Genetics/Occupational and Environmental Epidemiology Branch, plans to procure on a sole source basis services to perform a whole genome miRNA expression scan among exposed workers to diesel exhaust and exposed controls from Boston University 85 E. Newton Street, M-921, Boston, MA 02118. This acquisition will be processed under FAR Part 12 - Acquisition for Commercial Items and will be made pursuant to the authority in FAR Part 13.106-1(b)(1), using simplified acquisition procedures for commercial acquisitions. The North American Industry Classification System code is 541990 and the business size standard is $14.0 Million. Only one award will be made as a result of this solicitation. This will be awarded as a firm fixed price type contract. The period of performance is estimated to be twelve (12) months from date of award. It has been determined there are no opportunities to acquire green products or services for this procurement. Micro RNAs (miRNAs) are regulatory, non-coding RNAs about 22 nucleotides in length. The regulated expression of miRNAs may be important for cancer etiology. One miRNA can regulate multiple mRNA targets, and one target mRNA may be regulated by multiple miRNA sequences. Thus, the role of each miRNA sequence has yet to be fully elucidated. Evaluating miRNA expression profiles in cancer tissues may contribute to our understanding of miRNA-based gene regulation and cancer etiology. The NCI will evaluate the miRNA expression among workers exposed to diesel exhaust and controls. Diesel engines are used in automobiles, generators, light-duty and heavy-duty vehicles as well as railroad locomotives. When diesel fuel burns in an engine, the resulting exhaust is made up of soot and gases representing thousands of different chemical substances. 90% of the soot consists of <1um diameter particles that can be inhaled and deposited in the lungs. Diesel exhaust contains 20-100 times more particles than gasoline exhaust. These particles carry absorbed cancer causing substances such as nitro polynuclear aromatic hydrocarbons (nitro-PAH)'s. To compare the different characteristics of miRNA expression between exposed workers to diesel exhaust with unexposed controls, NCI will evaluate the miRNA expression in a diesel exhaust biomarker study in China. 1. Contractor Requirements The contractor shall perform the following tasks for 100 buccal scrape samples: 1. Log the receipt of biologic samples in a MS Excel file including the information of ID and date of receipt. 2. Perform whole genome miRNA expression scan. miRNA-seq libraries will be prepared from 200 ng of total RNA using the NEBNext® Multiplex Small RNA Library Prep Set for Illumina. Samples will be multiplexed at 10-12 samples per lane and sequenced on the Illumina HiSeq 2500 generating approximately 10-15 million 35-bp reads per sample. 3. Perform relevant data analysis on expression scan data. 4. Report problems in the analysis of samples to the Contracting Officer's Representative (COR) within three days when they happen. 2. Government Responsibilities NCI will ship 100 buccal scrape samples to the Contractor in one shipment. Samples will not have any personal identifiers attached. 3. Reporting Requirement and Deliverables The Contractor shall call the NCI Contracting Officer's Representative (COR) biweekly to report the number of samples processed. The Contractor shall deliver to the NCI's COR the expression sequencing datasets and results no later than September 10, 2017. 4. Inspection and acceptance criteria NCI's COR will have 15 days in which to review and accept these data. If no comments or request for revisions are provided within 15 days the deliverables shall be considered acceptable. Boston University has developed a unique sample collection method to noninvasively collect and preserve samples from the nasal airways and month for whole genome miRNA expression scan analysis. Boston University has applied this methodology to the analysis of upper airway samples collected from populations exposed to other air exposures that affect the lung and nasopharynx including previous research funded by the NCI Occupational and Environmental Epidemiology Branch. It is critical that data from previous NCI studies, including the analysis of mRNA, which this laboratory has performed on the diesel biomarker study, be scientifically comparable to the results of miRNA, to be supported under this procurement. Thus, Boston University is the only known source that can meet the Government's needs at this time. This notice is not a request for competitive quotation. However, if any interested party, especially small business believes it can meet the above requirement, it may submit a proposal or quote for the Government to consider. The response and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform the requirement. Responses must be received in the contracting office by 11 AM EST, on August 22, 2016. All responses and questions must be in writing and faxed 240-276-5399 or emailed to Reyes Rodriguez, Contracting Officer via electronic mail at reyes.rodriguez@nih.gov. A determination by the Government not to compete this proposed requirement based upon responses to this notice is solely within the discretion of the Government. Information received will be considered solely for the purpose of determining whether to conduct a competitive procurement. No collect calls will be accepted. In order to receive an award, contractors must be registered and have valid certification in the Central Contractor Registration (CCR) and the Online Representations and Certifications Applications (ORCA) through sam.gov.
