- The USDA, ARS, Western Regional Research Center, Albany, California has a requirement for one Nanoscale HPLC System, one LC/MS Peptide Calibration Kit, and one Nano Gradient Flow Module that combines the following features in one compact instrument: • Dual high pressure binary gradient pumps (100 nL/min- 50 µL/min), and additional isocratic (loading) pump (1-30 µL/min), allowing for a wide variety of applications including dual trap/dual column applications for increased sample throughput and on-line multidimensional separations for increased number of proteins identified. • Splitless (direct) pumps with flow monitoring and feedback to guarantee flow rate accuracy and reproducibility independent of backpressure and viscosity. • 10,000 psi max. pressure for both gradient pumps. Pump design with low mechanical and electrical noise that is required for proteomics/protein research. • 10,000 psi isocratic loading pump (1-30 µL/min) with flow monitoring and feedback. • Easy exchangeable flow modules for changing flow rate ranges, with all fluidic and electrical connections automatically made when replacing the module, whereby calibration data is stored without the need to recalibrate after switching flow ranges. • Flexibility to be reconfigured for ‘micro-flow' rates as high as 50uL/minute for higher throughput analyses. • Flow rate precision < 0.35% RSD at 500 nL/min. • Column oven with temperature ambient plus 5ºC to 60ºC. • Provide a single pump for isocratic sample loading, and two binary pumps for sample chromatography and chromatographic column washing which is necessary The nanoLC autosampler must provide the following features: • Programmable injection volume from 100 nL to 10 µL with 10 µL loop. • Precision of < 1% RSD for µL-pick-up loop injections > 1 µL. • Can be used with autosampler vials or microtiter plates. Capacity of two microtiter plates or 108-vial rack. • Bottom-of-vial detection for pick-up of small volume samples. • Dual needled design (Stainless Steel outer needle for septum piercing and PEEKsil inner needle for sample aspiration) allows for injection of small sample volumes. • One additional 10,000 psi 10-port column switching valve with the option to mount a second 10-port valve. • Sample cooling (4°C to 40°C). The nanoscale HPLC system software control and autosampler software control must be fully integrated with AB Sciex Analyst and Thermo Xcalibur Mass Spectrometry software (version 2.2.0.48) to allow direct interface with an existing Thermo Orbitrap Elite high resolution mass spectrometer that is equipped with a New Objective Picoslide nano-flow column switching device. System must provide a single pump for isocratic sample loading, and two binary pumps for sample chromatography and chromatographic column washing, which is necessary in utilizing the existing column switching apparatus. The pump software allows for recording of pressure read-backs from all channels (necessary for performance monitoring and system troubleshooting. The pumps are configured to run from 100-1000 nL/minute with a retention time precision of <1%). The maximum backpressure of 10,000 psi will allow use of state-of-the-art HPLC stationary phases maximizing chromatographic resolution and the mass spectrometer duty cycle. This system must also be compatible with Sciex Analyst software that is currently operating on an existing mass spectrometer, SCIEX Qstar Pulsar. Provide site installation, user manuals, warranties and shipping/freight fees. This solicitation incorporates the following commercial FAR clauses by reference 52.212-1, 52.212-2, 52.212-3, 52.212-4, and 52.212-5.
