This is a combined synopsis/solicitation for commercial items prepared in accordance with the Federal Acquisition Regulation (FAR) format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. The solicitation is being issued in conjunction with FAR PART 13.106. The solicitation number is FDA_16-233-1163080. This solicitation is issued as a Request for Quote (RFQ). The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) 2005-59-1, August 15, 2016. Requirement: The U.S. Food and Drug Administration (FDA), Center for Biologics Evaluation and Research (CBER), requires a Super-Sensitivity (Hybrid) Photodetector for an existing Leica SP8 Confocal Microscope System. Background: The FDA/CBER currently has an external single molecule detection system, which has limited capability due to the fixed emission filter wavelength and external slow machine response time. Researchers have to deal with cross talk and slow machine response time due to the external setup, which affect for sensitivity and accuracy of our assay. CBER has several ongoing Fluorescence Resonance Energy Transfer (FRET) measurement projects for vaccine and drug evaluation examination which are critical for FDA research programs. CBER analyzes the host-pathogen (e.g. virus, bacteria, parasites) interaction using cultured cell and tissue in immunological responses and vaccine related studies. Statement of Need: The Photodetector is required for fluorescently labeled cell sample image acquisitions and spectral Fluorescent Lifetime Imaging Microscopy (FLIM), spectral Fluorescence Correlation Spectroscopy (FCS), Fluorescence Cross-Correlation Spectroscopy (FCCS) and Fluorescent Lifetime Correlation Spectroscopy (FLCS) to measure fluorescence energy transfer and molecular mobility in single molecule level. The Super-Sensitivity Photodetector is required with high sensitivity enabled to eliminate the cross talk between the dyes with faster response time for single molecule detection on weak signals which is biologically important for research. The system will be used to visualize and evaluate samples in super resolution using gated Stimulated Emission Depletion (STED) microscopy technology for samples from a variety of experiments targeted towards understanding the cellular consequences, to evaluate the vaccine or drug candidates, and evaluate efficacy of agents to viruses or bacteria in a variety of species and cell types for our researchers. Minimum Technical Specifications: - Shall be compatible with, attached to, and work seamlessly with an existing Leica SP8 Confocal Microscope System (S/N 8100000213) - Shall have two hybrid single molecule detectors with external cooling unit for single molecule detection to reduce back ground noise to obtain the better image and data - Shall have capability of internal single molecule image and data acquisition such as spectral FLIM, FCS, FCCS and FLCS to avoid cross talk with rapid data sampling with high resolution - Shall have single photon counting mode for detection - Shall work with gated STED microscopy for super resolution microscopy - Shall have external cooling circuit - Shall acquire molecular mobility, binding, viscosity and kinetic data from our various samples using 4 channel FCCS technology to look interaction of multiple molecular binding partners in live samples with fewer artifacts - Shall have pulse interface for synchronization of Time-Correlated Single Photon Counting (TCSPC) module and scanner - Shall have communication interface between hardware control software and detectors - Shall have fast (<130ps) hardware response time for high quality FLIM data acquisition including FLIM-FRET (Forester Resonance Energy Transfer) efficiency analysis. - Shall have 64 GB RAM workstation for fast spectral FLIM, spectral FCS and FLCS image /data acquisition and analysis on 64 bit SMD workstation on current SP8 system - Shall have offline dongle for data analysis - Shall be delivered with all necessary equipment, components, adapters, software, accessories, cables, wheels, start-up supplies, etc. - Shall provide Set-up and Installation - Shall demonstrate upon installation that the item meets or exceeds all performance specifications. - Shall provide on-site operator training/familiarization to include system operations, calibration, optimization, trouble-shooting, and basic operational maintenance procedures for at least four (4) attendees Alternative solutions to providing the same upgraded capabilities as described above will be considered. Solicitation provisions- The provision at FAR 52.212-1 Instructions to Offerors - Commercial Items applies to this solicitation. The following FAR 52.212-1 addenda apply: Paragraph (b)(4). To ensure information is available, offerors shall furnish as part of their proposal all descriptive material necessary for the government to conclusively determine that the product offered will be technically capable of meeting or exceeding the Government's requirements. Offeror shall further demonstrate (i.e., published reports, scientific articles, etc.) that the proposed solution has been proven to work seamlessly with an existing Leica SP8 Confocal Microscope System The government is not responsible for locating or securing any information, which is not identified in the proposal. Paragraph (k) To be considered for award an offeror shall be registered and active in SAM by the date and time set forth for receipt of quotes/proposals. The provision at FAR 52.212-2 Evaluation-Commercial Items applies to this solicitation. The following subparagraph (a) factors apply: (i) Technical Capability (See FAR Provision 52.212-1 (b)(4) and addenda thereto) (ii) Price. Technical Capability (i.e., how well the offered product meets or exceeds the Government's requirements) is more important than price in determining the best value to the Government though price remains a significant consideration. The Provision at FAR 52.212-3, Offerors Representations and Certifications-Commercial Items, applies to this acquisition. An offeror shall complete only paragraph (b) of the provision if the offeror has completed the annual representations and certification electronically via https://www.sam.gov/portal/public/SAM/#1. If an offeror has not completed the annual representations and certifications electronically at the website, the offeror shall complete only paragraphs (c) through (o) of this provision. It is the offeror's responsibility to monitor the internet site for the release of an amendment to the combined synopsis/solicitation (if any).
