Description
This is a Full & Open Notice for brand name or equal Hybrigenics ULTimate Y2H and Y2H+1 in accordance with the format in FAR Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals that is being requested and a written solicitation will not be issued. Solicitation No. AG-5615-S-17-BB01 is issued as a Request for Proposals (RFP). This solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2005-26. The NAICS code applicable to this acquisition is 541711. The USDA, ARS, MWA, Vegetable Crops Research Unit, Madison, WI has a need for a screen for protein interactors using a yeast-based system with the ability to search for direct interacting proteins (yeast 2-hybrid) as well as the ability to search for interacting proteins when a third protein of interest is present (yeast 3-hybrid). The requirement is for at least 20 genes from Nicotian benthamiana that encode proteins and interacts with the Phytophthora infestans effector protein IPI-O1 in the absence of the Solanum tuberosum protein RB, and at least 20 genes from Nicotiana benthamiana that encode proteins that interact with IPI-O1 in the presence of RB. This system will be used to identify plant proteins that interact with the Phytophthora infestans effector protein IPI-O1. This effector is required for infection of potato and other host plants. The corresponding resistance protein from potato, called RB, recognizes the presence of the IPI-O1 effector to elicit resistance. We require additional information about the function of IPI-O1 in the infection process, including what host proteins it interacts with. This will give us an idea of what its targets are and, therefore, an idea of how it is used to cause disease. THE SALIENT CHARACTERISTICS: The Nicotiana benthamiana system must meet these minimum requirements: 1) Have access to an existing cDNA library from Nicotiana benthamiana, 2) Be able to complete a screen to identify direct interactions between a single bait protein and a library of putative interacting proteins, 3) Be able to complete a screen to identify direct interactions between a single bait protein and a library of putative interacting proteins in the presence of a third protein, 4) Use a yeast mating protocol to screen a large number of individual clones, 5) Be able to screen at least 10 million independent gene fragments, 6) Able to test all protein domains for interaction with the bait protein, 7) Sequence and analyze all clones identified in the screen, 8) Provide a comprehensive bioinformatics analysis of identified interacting proteins, 9) Provide a confidence score with each interacting protein. References must have proposed research within the last three (3) calendar years. PROPOSAL PRICE MUST INCLUDE: shipping or handling fees, as well as any applicable duties, brokerage, or customs fees. DELIVERY DUE DATE: The Government requires delivery within three (3) months after award and PO is issued, however, each offeror shall include their proposed delivery schedule as part of their quote. FAR 52.212-2, EVALUATION - COMMERCIAL ITEM, the significant evaluation factors, in the relative order of importance, are (i) technical capability of the item offered to meet the Government requirement including delivery date; (ii) past performance; and (iii) price (based on F.O.B. Destination), technical and past performance are more important than price. Award will be made based on overall BEST VALUE to the Government. EVALUATION & AWARD FACTORS: The contract resulting from this solicitation will be awarded to the responsible party whose offer, conforming to the solicitation, is determined most advantageous to the government, cost, and other factors considered. Each Proposal shall respond to each of the evaluation factors identified below. With the exception of past performance, a technical evaluation will be performed on each quote based solely on the information furnished and not on previous knowledge or associations.
