Description
This contract action is for the acquisition of services for which the Government intends to solicit and negotiate with only one source under the authority of Federal Acquisition Regulation (FAR) Subpart 13.106-1(b)(1). The National Institute of Environmental Health Science (NIEHS), Office of Acquisitions (OA) intends to award a single source purchase order to Active Motif, 1914 Palomar Oaks Way, Suite 150, Carlsbad, CA 92008, for the acquisition of services to Evaluate Mouse Uterine Chromatin Landscaping. The Receptor Biology Group investigates estrogenic responses in multiple organ systems and the role of estrogen receptors and gene networks in these responses. The group studies the cellular mechanisms of estrogen receptor activation and the structural and chemical basis for the stimulation of estrogenic responses by endogenous and environmental compounds. The Receptor Biology Group requires Chromatin ImmunoPrecipitation Seq (ChIP-seq) of 2 samples (Vehicle or 1h Estrogen treated mouse uterus) to evaluate the cohesion complex and determine possible looping interactions that are suggested by our previous observation of numerous ERa binding sites that are 50-200 KB distal from regulated transcripts. The Receptor Biology Group also requires ChIP-PCR experiments to verify findings from previous experiments as well as the proposed one (from WT and Igf1 enhancer knockout uterus treated with: vehicle, 1h E2, 1h IGF1 or 1h ICI for a total of 8 samples) using 5 different antibodies to evaluate ERa, RNA polymerase II, cohesion complex, and two different Histone H3 modifications (K27acetyl, K4trimethyl) and primer sets in six different regions to verify binding. NIEHS will furnish to the contractor the biological samples for analysis along with a listing of samples by date and subject identification number and instructions on which chromatin binding factors are to be analyzed. In some cases antibodies will be supplied by NIEHS. Required capabilities: • Cross-link transcriptional proteins to chromatin within the tissue we provided • Fragment and isolate the chromatin • Enrich the chromatin for estrogen receptor alpha (ERα), RNA polymerase II (PolII), p300 or cohesin using immunoprecipitation • Perform PCR to detect co-precipitated DNA regions (ChIP-PCR) and provide us with the data OR • Construct libraries from the enriched chromatin for next-generation sequencing • Sequence all the chromatin DNA in the immunopreciptated/enriched chromatin using Next-generation sequencing • Provide us with the data detailing all the sequences that interact with ERα, PolII, p300 or cohesin in each sample It has been determined that Active Motif, is the only known responsible source capable of providing these supplies and no other service will satisfy the agency's requirements. The ability to maintain continuity of experimental procedures is crucial to the success of the Division of Intramural Research, Receptor Biology Group, at the NIEHS. The applicable North American Industry Classification System (NAICS) Code for this acquisition is 541711 "Research and Development in Biotechnology" and a Size Standard of 1000. The anticipated period of performance is September 30, 2017-September 29, 2018. NIEHS will solicit and negotiate directly with Active Motif and no solicitation will be issued. This notice of intent is not a request for competitive proposals or quotations; however, all responsible sources may submit a bid, proposal, or quotation which shall be considered by the agency. Offerors must have an active registration in the System for Award Management (SAM) www.sam.gov. A determination by the Government not to compete this proposed requirement based on responses to this notice is solely within the discretion of the Government. Interested parties must express their interest and must be submitted in writing via email to Valarie K. Sims at valare.sims@nih.gov and are due by 10:00 AM, August 11, 2017. No faxed documents will be accepted. If no responsive statements are received by the due date, an award will be made to Active Motif.
