Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NIH-NIDDK-17-216 and is issued as a Request for Quotation (RFQ). The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2005-95. The North American Industry Classification (NAICS) Code is 334516 and the business size standard is 1,000. However, this solicitation is not set aside for small business. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) intend to procure on a sole source basis with BIOREP TECHNOLOGIES. of 15804 NW 57TH AVE MIAMI LAKES FL 33014 for the purchase of an AUTOMATED PERIFUSION SYSTEM WITH VALVE and manual tray handling. The National Institute of Digestive, Diabetes & Kidney Diseases's (NIDDK) requires an automated perifusion system to stimulate cells or tissue with solutions and measuring the hormones excreted. This machine will allow continual measurement of secreted peptides and molecules from cultured primary cells and cell lines. We expect to use this for multiple types of glucose and fatty acid responsive primary cells (islets cells of the pancreas, hepatocytes, adipocytes, muscle cells). Other agents like hormones and cytokines (insulin, TGF-beta, leptin, exendin-4/GLP1, short-chain fatty acids like butyrate) can also be assayed for their cellular responses in this system. The description for experiments using cultured rodent and human islets follows. The NIDDK anticipates that this system can be similarly used to study the stimulus-coupled secretory properties of other cells (adipocytes, hepatocytes, muscle cells). The lab and branch has access to many transgenic and genetically engineered cell/tissue-specific conditional mouse models that can be utilized to harvest the target cell type to address a specific question. Assays that involve measurement of insulin secretion in static incubation for 1 hour in buffers containing varying glucose concentrations do not provide an insight into minute-to-minute secretory profile of islet beta cells. In addition, with routine static incubations, assessment of insulin secretion to a glucose ramp over time is not possible. The BIOREP TECHNOLOGIES perifusion system can be used to assess dynamic insulin secretion from isolated islets in a physiologically relevant way. In addition, regulation of first and second phases of insulin secretion together with sustainment of insulin secretion can be studied in detail. While static incubation method is the simplest type of assay, it is a rudimentary analyses of insulin release which is dynamic and pulsatile depending on the stimulus used (glucose, fatty acids, GLP1, etc). Perifusion of islets is the most effective method in understanding dynamics of insulin secretion under conditions of physiological adaptation and pathologic cellular stress. There are no alternative or similar machines on the market. Manual and home-grown alternatives do not provide the breadth or depth of scientific data produced by this system. This equipment will aid thorough investigations of hormone secretion dynamics from a variety of cell types including pancreatic islet beta cells and adipocytes. The proposed contract action is for supplies or services for which the Government intends to solicit and negotiate with only one source under authority of the 41 U.S.C. 253(c) (1), FAR 6.302. Only one responsible source and no other supplies or services will satisfy agency requirements. The offeror must include a completed copy of the provision of FAR Clause 52.212-3, Offeror Representations and Certifications - Commercial Items with its offer. The provisions of FAR Clause 52.212-1 Instructions to Offerors - Commercial Items; FAR Clause 52.212-2, Evaluation - Commercial Items; FAR Clause 52.212-4, Contract Terms and Conditions - Commercial Items; and FAR 52.212-5, Contract Terms and Conditions Required to Implement Statutes or Executive Orders - Commercial Items - Deviation for Simplified Acquisitions applies to this acquisition. The offeror must include their Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size.
