Description
Notice of Intent: The USDA, ARS, SEA intends to award a sole source procurement to Promega Corporation, 2800 Woods Hollow Road, Madison, WI 53711 USA as they are the sole source for this type of multi-channel RNA / DNA extraction robot pursuant to the authority of 41 U.S.C. 253(c)(l ) and FAR 6.302-1, Only One Responsible Source and No Other Supplies or Services Will Satisfy agency Requirements. The award will be for a 48 placement cartridge robotic extraction system able to perform low volume (≤ 50µL) nucleic acid extraction from multiple insect tissue types without requiring any supplementary unique or proprietary commodities or software and which can seamlessly interface into our existing diagnostic platform, allowing multiple investigators to maintain the integrity of existing archived experimental sample and data sets. These low volume elutions of concentrated high-quality RNA must be able to be utilized immediately for use in downstream applications such as quantitative qRT-PCR and sequencing, with no post-purification concentration required. The unit should be supplied with diagnostic kits in prefilled cartridge format to maximize simplicity and convenience and also to eliminate possible buffer formulation or cartridge loading errors. The sample preparation protocol should have minimal operator required steps, preferably not more than four, and should not require any centrifugation or filtration components. This serves to reduce the processing time and minimize the potential for cross-contamination of samples and limits other prospective lab-induced sources of error. The nucleic acid yields must maintain the highest integrity, consistency and reliability. Each cartridge should have the ability to extract from up to 20mg of tissue, as pooled sample processing is becoming a common requirement in our application. The extraction protocol should NOT require the use of β-mercaptoethanol (BME) nor the use of any hazardous organic solvents, such as xylene or phenol, ensuring a minimal amount of hazardous waste is generated and thereby ensuring a greater level of user safety than other common methods. The system should also allow for integration of a fluorometer for sample quantitation that is capable of consolidating purification and nucleic acid quantification data into a single report. The system should function on the principle of being a magnetic particle mover, not a liquid handling system, thereby creating a minimal risk of cross-contamination due to the prospects of liquid handling errors or splashing during sample processing. The unit must also be able to be used optimally WITHOUT proprietary sample processing chemistries and with samples that do NOT homogenize thoroughly prior to being placed for extraction, yet still maintain the highest quality nucleic acid yields immediately suitable downstream applications. Promega Corporation is the sole source for this manner of extraction system that can satisfy all the requirements of the multiple investigators who utilize this method of analysis for their projects. The Government intends to award a firm fixed price Purchase Order on or about August 15, 2017, using Simplified Acquisition Procedures. This Notice of Intent is not considered a Request for Quotation (RFQ) nor is it a Request for Proposals (RFP). No solicitation document is available and telephone requests will not be honored. Interested persons may identify their interest and capability to respond to this requirement. A determination by the Government not to compete this proposed contract based on responses to this notice is solely within the discretion of the Government. The Government will consider all responses received by 11:OO AM central time, August 14, 2017.
