Description
INTRODUCTION The Veterans Health Administration (VHA), Program Contracting Activity Central (PCAC) anticipates awarding a Sole Source, Firm-Fixed-Price contract for a Protein Gel Electrophoresis Workflow and Documentation System for the Activation of the Research and Development at Puget Sound Healthcare in Seattle, WA. Acquisition of this item is part of the initial outfitting and activation of the new VA Medical Center being constructed in Seattle, WA DESCRIPTION A Justification and Approval has been established based on Market Research conducted by the Department of Veterans Affairs. The Contractor shall deliver all items. All items must meet all salient characteristics and the contractor shall furnish all personnel, supplies, and equipment required for delivery. This requirement is a brand name and sole source justification for all CLINs outlined in the Salient Characteristics below. Salient Characteristics CLIN 1 Packaged system: Gel workflow and documentation with consumables Basis of Design BIO RAD ChemiDoc MP V3 Western Workflow Midi Gels Qty: 1 (Bio-Rad# 17001389) Gel documentation System: Unit will collect relevant data without integration or connection to the VA LAN / network High signal to noise scientific camera comparable to or better than a 6 mp scientific supercooled 16-bit data acquisition CCD camera System requires a combination of high-resolution imaging and sensitivity 6.45 × 6.45 m pixel size to resolve closely spaced bands on a western blots or gel as well as for longer exposures Peltier cooling to 30 °C, absolute and regulated integration time for weak chemiluminescent signals System must have 4+ orders of linear dynamic range System must be able to accommodate multiple imaging modalities / signal types to include: Trans-UV: EtBr, SYBR ® Green, SYBR ® Safe, SYBR ® Gold, GelGreen, GelRed, (for the fluors listed but not limited to) Fluorescein, OliGreen, PicoGreen, GelStar, Stain-Free, Oriole, SYPRO Ruby, Pro-Q Emerald 300, Epi-blue: Coomassie Fluor Orange, Alexa Fluor 488, Dylight 488, Pro-Q, Emerald 488, QDot 525, QDot 605, QDot 625, QDot 705, Epi-green: Flamingo, Krypton, Pro-Q Diamond, Cy3, Alexa Fluor 546, Dylight 549, Rhodamine, Epi-red: Cy5, Cy5.5, Alexa Fluor 647, Alexa Fluor 680, Dylight 650, Dylight 680 Epi-far red & near infra-red: IRDye 680 & IRDye 800 Trans-white: Coomassie stain, silver stain No light imaging chemiluminescence System should include a source of image analysis documentation and quantitation software for both Windows and Apple based computers. System should have unrestricted user copies of Image analysis software System must have preset protocols for proprietary Stain-Free technologies allowing for optimized image acquisition for gels and blots to eliminate coomassie staining and/or Ponceau S staining of gels and blots. System should have access to future installations and free upgrades to new versions of analysis and documentation software. System must yield reproducible image data without any imaging artifacts System should provide automated multiplex image analysis for Total Protein Normalization to eliminate the requirement to show loading control proteins on Western blots System should provide total protein normalization with stain-free technologies, if possible, as an alternative to normalization with single housekeeping proteins (HKPs). System should provide automated multichannel image viewing and color-coded analyses in during acquisition to generate multiplex image files for easy downstream use System must have automatic focus and iris adjustments for image acquisition System should provide flexible and automated image exposure setting for all applications to ensure optimal exposure times depending on user needs. System should provide 16-bit and 8-bit tiff images with a one-click export options, allowing users to retain all image data as a 16-bit or 8-bit.tiff file. System should provide publication-ready images with a one-click export option in resolutions, formats and dimensions to be specified. System should provide for automatic generation of customizable reports System should be able to produce customizable reports with data organized as desired, including lane and band identification, molecular weight or base pair evaluation. Band sizing and quantification to be based on reference bands or quantity standards. System should provide large descriptive buttons with tool tips and comprehensive tutorial within navigation menus Blot/UV/Stain-Free Sample Tray(s) Basis of Design BIO RAD Qty: 1 black surface tray (Bio-Rad# 12003028); 1 white sample tray (Bio-Rad # 12003026); 1 blue sample tray for GelGreen and or SYBR Stains (Bio-Rad# 12003027) sample tray(s) for imaging chemiluminescence and fluorescence blots, stain-free gels and blots, and gels stained with ethidium bromide, SYPRO Ruby, Oriole, or SYBR ® Stains Tray(s) must accommodate the imaging of a wide range of different gels and blots A single tray that can accommodate all required imaging modalities listed in CLIN1 (g) is acceptable; this requirement is for functionality, not a specific number of sample trays The tray(s) must be supplied with the gel documentation system or approved by the gel documentation system s manufacturer for the visualization of the following: Bio-Rad stain-free gels and blots Single and multiplex fluorescence blots (RGB, far red, near IR) Chemiluminescence Gels and blots stained with ethidium bromide, SYPRO Ruby, Oriole, GelRed and SYBR ® Stain, and other dyes C) Semi-Dry Transfer system Basis of Design BIO RAD Qty: 1 (Bio-Rad# 1704150) Must have demonstrated 3 minute, 7 minute, 10 minute and user-defined transfer protocols of proteins from PAGE gels to nitrocellulose, LF-PVDF or PVDF membranes Must have demonstrated successful transfer of at least 1.25 ng protein to membranes with documented detection of protein bands using ECL-based detection reagents Must allow up to accommodate the transfer of at least 2 midi gels simultaneously Must allow the assembly and running of transfers independently System must accommodate both traditional semi-dry and well as rapid transfers System must be designed to accommodate use of ready-to-use filter paper + membrane prepackaged consumables to reduce set up times, improve consistency and reduce user error System must come preprogramed with optimized preloaded protocols and allow customization and save/recall of user-defined protocols System must have demonstrated uniform transfer of high, medium and low molecular weight proteins as determined by published low intra-blot CV near 9% or less. System must provide superior protein transfer efficiency as demonstrated by 2-D spot quantitation when compared similar systems System must come with a total 5-year service warranty or be covered under an equivalent warranty associated with the package purchase of items in this requirement D) Poly acrylamide gel electrophoresis systems for protein gels Basis of Design BIO RAD Qty: 1 (Bio-Rad # 1656001) Vertical midi-format electrophoresis cell, includes buffer tank, lid with power cables, 3 sample loading guides (12+2-well, 18-well, and 26-well) System must be compatible with the power supply unit as described in the Salient Characteristics section below Must be able to accommodate Criterion TGX precast gels of midi size that are offered by BioRad. Must be constructed of clear plastic Have UV transparent gel trays with built in fluorescent ruler to determine band migration lengths Must be able to accommodate 2 gels simultaneously.
