- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in FAR 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation and a separate written solicitation will not be issued. This solicitation number is NICHD-17-288 and is issued as a Request for Quotation (RFQ). The solicitation/contract will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2005-95. The North American Industry Classification (NAICS) Code is 541712 and the business size standard is 1000 employees. However, this solicitation is not set aside for small business. This acquisition is being conducted using Simplified Acquisition Procedures in accordance with FAR Part 13. Eunice Kennedy Shriver National Institute of Child Health and Human Development (NICHD) intend to contract on a sole source basis with Metabolon, PO Box 110407, Research Triangle Park, NC 27709 for Analysis of metabolomics lipids and short chain fatty acids in 24 plasma samples and 24 fecal samples of mice with different microbiome. The NIDDK, NIH is studying the effect of the microbiome on host physiology and immune responses. To this end, investigators in the section have found that the gastrointestinal microbiome affects systemic immune responses and that it alters resistance to colorectal carcinogenesis and viral infection. To understand whether effects of the microbiome on host metabolism contribute to these observations, we are planning to analyze metabolites, lipids and short chain fatty acids in plasma samples and in stool samples of three groups of mice (8 mice per group) that differ in their microbiome (24 plasma samples and 24 stool samples total). Scope of Work/Description of Proposed Tasks to be Performed Task 1: Analyze and identify metabolites in plasma samples and in stool samples of three groups of mice (8 mice per group) that differ in their microbiome (24 plasma samples and 24 stool samples total) by liquid chromatography/tandem accurate mass spectrometer. Samples should be divided into 4 aliquots, one of which should be analyzed by UPLC-MS/MS with positive ion mode electrospray ionization, one by UPLC-MS/MS with negative ion mode electrospray ionization, one by LC polar platform, and one by GC-MS, Each detected biochemical should be compared to a library of at least 4000 named compounds, using retention index and mass spectrum. Provide quantitative comparison of the amount of these metabolites in the plasma and stool samples of the different mice. Task 2: Analyze and identify lipids in plasma samples and in stool samples of three groups of mice (8 mice per group) that differ in their microbiome (24 plasma samples and 24 stool samples total) by differential mobility spectroscopy (DMS). Each detected lipid species should be compared to a reference data with masses from at least 1000 lipid species. Provide quantitative comparison of the amount of these lipid species in the plasma and stool samples of the different mice. Task 3: Analyze and identify short chain fatty acids in plasma samples and in stool samples of three groups of mice (8 mice per group) that differ in their microbiome (24 plasma samples and 24 stool samples total). Provide quantitative comparison of the amount of these metabolites in the plasma and stool samples of the different mice. Task 4: Assign the list of detected metabolites, lipids and short-chain fatty acids to specific biochemical pathway with appropriate statistics. Provide quantitative analysis (absolute, relative amount and heatmap) and statistical comparison between the findings for the three groups of mice. Metabolon has also recently analyzed samples from patients with liver diseases collected by other investigators in the NIDDK. Because investigators in theNIDDK were involved in the immune response analysis of these patients, the use of the same vendor Metabolon for the here proposed study would allow a direct comparison of the data sets from both studies, which may aid data interpretation and conclusions.
