- This is a combined synopsis/solicitation for the United Stated Department of Agriculture, Agriculture Research Service located in Beltsville, Maryland 20705 - for the following commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation, quotes are being requested and a written notice will not be issued. Solicitation 880629 is issued as a request for quotation (RFQ). The solicitation document and incorporation provisions and clauses are those in effect through Federal Acquisition Circular 2005-95. This solicitation will be reserved exclusively for small businesses with a small business size standard of 1,000 employees referencing North American Industrial Classification Code. (NAICS) code 334516. The following requirement: Brand Name or Equal to, PipetmaX Automated Liquid Handler instrument - PipetmaX with cover cutouts for component placement, Quantity of one each. Minimum requirements: Genome and metagenome sequencing as well as PCR, qPCR, and antibiotic sensitivity assays are important aspects of our research that require repeated precise pipetting. Due to the amount of pipetting time required for these assays as well as the potential human error that is inherent in manual pipetting we would like to purchase an automated, robotic liquid handler that can perform these functions on a laboratory bench top, as well as inside of a biological safety cabinet. When USDA sequences bacterial genomes, USDA prepares 384 isolates for an individual sequencing run, the protocol that USDA uses to prepare these isolates shall provide at least 1680 pipetting actions with an 8-channel pipet, and shall provide 1536 pipetting actions with a single channel pipet, and shall provide 1540 volume adjustments. Obtaining high-quality DNA sequence requires precise and accurate liquid transferring technique, therefore automation will improve the quality of generated data. Conventional PCR and qPCR are routinely performed in USDA's lab and these protocols shall require precise and accurate liquid transferring techniques. The following are the (minimum requirements ) of the required liquid handler that shall be required to meet USDA research needs.: The liquid handler shall be used for preparing bacterial DNA libraries for sequencing. This procedure is generally carried out on the open laboratory bench. The liquid handler shall also be used to prepare RNA sequencing libraries and to dispense/transfer bacterial pathogens. The above two procedures shall be done in a biosafety cabinet. Therefore, the liquid handler shall be used in two different locations and the system shall be light enough, less than 60 pounds, to be installed and moved by one to two lab USDA technicians and shall be small enough to fit in one half of the biosafety cabinet, and shall be no more than 33 inches x 22 inches x 22 inches, in Width, Diameter and Height. Sideways is accepatable. The instrument shall have the ability to be set up for multiple applications: library prep, PCR prep, sensitivity plate prep and shall be easily and quickly reconfigured for application changes. The above required liquid handler shall be compatible with common molecular biology labware and shall be Compatible with SLAS- ANSI footprint 96-well plates, SLAS-ANSI footprint 384-well plates, SLAS- ANSI footprint deep reservoirs, 1.5ml tubes, 2.0ml tubes, and PCR strip tubes. The liquid handler shall be able to load any number of tips on its multichannel pipette heads. The liquid handler shall be able to transfer liquids in the range of 1uL to 200uL of volume from one well of 96 or 384 well plate to another well in another 96 or 384 well plates, with variable volume as well as same volume from well to well. The above required system shall allow the USDA End-user to set up labware for a run, while the instrument is performing a different run. The liquid handler shall be capable of providing at least two different pipetting heads in a single method without manual intervention to maximize the effective volume range. The liquid handler shall perform with the same accuracy and precision as a high quality manual pipette which USDA already purchased from the manufacturer Gilson. Maximum permissible random error at 1μl, 5% Maximum permissible systematic error at 1μl: 8%. The liquid handler shall be capable of reverse pipetting and repetitive pipetting. Because USDA has multiple end-users, the liquid handler shall have control software that is easy to operate to allow users to be up in running the system with minimum time in training. Additional items required to operate the liquid handler: PipetmaX Riser Kit a. The required liquid handler shall allow large numbers of tips to be ejected and stored away from labware being used in the method running on the instrument. b. PipetmaX 8 x 200 Pipette Head. The liquid handler shall have the capability to pipette up to 200 ul in 8 wells per event c. PipetmaX 8 x20 Pipette Head The liquid handler shall have the capability to pipette 20 ul in 8 wells per event d. PipetmaX 384 Compatible Tray The liquid handler shall have the capability for operating with multiple plate configurations of. 96 or 386 well plates. e. PipetmaX Tip reload Block The above item is required as a place to load the pipet tip container. f. 1.5-2.0 ml Flip Cap Tube Rack Code 424 The above item is required for holding reagent tubes or sample tubes. g. PCR Tube Rack Code 496 The above item is required for holding sample tubes. h. NORMALIZATION ASST LICENSE, LIFETIME The above item is required for the software programing. I. SOFTWARE PROTOCOL BUILDER V1 The above required software shall be user friendly and shall have the following capabilities: Shall have the Capability to import and store multiple protocols. Shall have the Capability to accommodate variables for parameters to be satisfied at run time. Shall provide Graphical instrument setup guidance including labware positions, solution positions and instrument components. Shall provide Graphical tracking of method. Shall provide Real time tracking solutions and volumes transferred. Shall have the Capability to label wells at runtime and track their contents throughout a method. Shall provide the Capability to define initial volumes at run time. Shall provide the Capability to transfer data, methods, reports via USB stick from the instrument itself. Shall provide the capability to simulate a method before running and view a virtual representation of the run. Shall provide the Capability to control RS-232 and Ethernet devices as part of a method run file. Shall provide the Capability to calculate liquid level throughout a method and determine aspirate-dispense heights based on it. Shall provide the Capability to save and view results files after a run, including error reports and solution locations. j. PipetmaX STANDARD TABLET Kit V2 The above item is required as an interface for communicating with the liquid handler, collecting data, and transfer of data. k. INSTALLATION/FAMILIARIZATION FOR 1 DAY INSTALL l.
