THE CORRECT SOLICITATION NUMBER SHOULD BE 1232SD18P0420; NOT 1232SD18P0425. ALL OTHER INFORMATION & DUE DATE REMAIN UNCHANGED. The USDA ARS - WBSC intends to award a sole source Firm Fixed Purchase Order (PO) to UNIVERSITY OF NORTH CAROLINA AT CHAPEL HILL (UNC), 104 AIRPORT DRIVE, CB# 1350, CHAPEL HILL, NC 0262-4658. This Order is to provide continuous research support services from UNC Histology Core Laboratory (Core), processing tissues provided by USDA ARS Scientists for the ongoing research project at USDA ARS, Western Health Nutrition Research Center (WHNRC), 430 West Health Sciences Drive, Davis, CA 95616. The Firm Fixed PO will include the following: 1. SECTIONING OF FROZEN SAMPLES: SECTIONING OF TISSUE & MOUNTING ON MICROSCOPE SLIDES; 289 HRS; 2. MASSON'S TRICHROME STAINING; 12 HRS; 3. OIL O RED STAINING: 12 HRS In support of the Animal Atherosclerosis Studies as presented in American Heart Association (AHA) Statement, "Arteriosclerosis, Thrombosis, and Vascular Biology (ATVB), http://atvb.ahajournals.org/, USDA ARS, WHNRC, along with the collaborating efforts of UNC Histology Core Facility (Core), has strived to perform the best research as recommended in AHA Statement. UNC Core has played a very supportive & instrumental role in WHNRC research project by processing all the other specimens related to this project, and thus, it's critical that they finish the work to ensure continuity & comparability across the many samples of this project. A change at this point would be catastrophic to WHNRC research and may prevent them from meeting the goals and deadlines of the NIH funded research project As result and to maintain the stated continuity, comparability & consistency of their current research project, WHNRC requires the continuum support services & efforts of UNC Core in the histological processing of tissues as follows: • Ten (10) micron serial interrupted cryosectioning of mouse hearts eight (8) sections per slide, ten (10) slides per mouse. • The area of interest is the aortic values and ascending aorta. • One (1) slide is stained with Masson's Trichrome and one (1) with oil red O. • Knowing exactly where to start and stop, serial interrupted sectioning requires the technician to put one (1) section on each of the ten (10) slides and then section 11* goes into position 2 of slide 1 and this is repeated throughout the tissue. *Section11 - they section every ten (10) microns for 80 section total. UNC Core, a 600-square foot research laboratory, offers a variety of services insuring that histological techniques are always available for investigators and their research project needs; and is equipped with two Leica 1950 cryostats, a Cryojane Tape Transfer frozen sectioning system, two Leica 2235 rotary microtomes, microscopes and a Vibratome. The Core has the equipment and expertise to produce reliable high-quality sections from fixed and unfixed tissues, frozen and paraffin-embedded tissues. Furthermore, they provide a unique serial interrupted sectioning technique. This technique allows analysis of the full length of 4mm vessels or tissues, collected on multiple slides with each slide containing serial sections representing a specific distance through the full length of that tissue. In addition, special stains and immunohistochemical assays are routinely performed at the Core. The Core offers high quality, reliable, cost effective histological services including but not limited to: Tissue Sectioning: Paraffin embedding and frozen embedding in OCT • Paraffin section - serial, random, serial interrupted • Frozen section - thaw mount cold/warm, serial, free floating, thick slice, random and serial interrupted, CryoJane tape transfer • Vibratome section Staining: • Routine staining, i.e., H+E, Nuclear Fast Red • Special staining, Alcian Blue, Cresyl Violet, PAS, Oil Red "O", Toluidine Blue, Von Kossa, Combined Massons Elastin, Picrosirius Red, Sudan IV, Thionin, Prussian Blue, Movats Pentachrome, Masson Trichrome, Alcian Blue/Phloxine/Tartrazine, others available upon request. Immunohistochemistry/Enzyme: • ABC method (direct/indirect) • Fluorescence • Biocytin detection • HRP • Single and multiple-labeling • Edu/Brdu in situ Hybridization • RNAScope • BaseScope Pursuant to the authority of 41 U.S.C. 253(c)(1) and FAR 6.302-1, Only One Responsible Source and No Other Supplies or Services Will Satisfy Agency Requirements. The award date will be on or about July 20, 2018; and this Notice of Intent is not considered as a Request for Quotation (RFQ) nor is it a Request for Proposals (RFP). No solicitation document is available and telephone requests will not be honored. Interested persons may identify their interest and capability to respond to this requirement. A determination, by the Government, to compete this proposed PO based on responses to this notice is solely within the discretion of the Government. The Government will consider all responses received by 10:30AM PST, July 19, 2018. Information received will be considered solely for determining whether to conduct a competitive procurement.
