- Please note that this is NOT a request for quotes or proposals, the Government is seeking information for market research purposes only. VHA Reginal Procurement Office (RPO) 2 is currently conducting a market survey for qualified firms capable of providing Optima XE equipment. The North American Industry Classification Systems (NAICS) is 334516. Responses to this notice must be submitted in writing (email or fax) and must be received no later than July 12, 2019 3PM EST. Interested parties MUST provide company/individual name, a capability statement, examples of same or similar work performed at other facilities, DUNS number and address, point of contact and social-economic. Interested parties who do not submit the above information on or before the response due date and time will not be considered for this procurement. Please review the attached Statement of Work attached herein. The purpose of this notice is to gain knowledge of potential qualified sources and their size classification/socioeconomic status (service-disabled veteran owned small business, veteran owned small business, women owned small business, HUB Zone, 8(a), small business or large business), relative to NAICS 334516. Responses to this notice will be used by the Government to make appropriate acquisition decisions. A solicitation is not currently available. If a solicitation is issued, it will be announced on GSA E-buy website http://www.fbo.gov at a later date, and all interested parties must respond to that solicitation announcement separately from the responses to this announcement. Statement of Work-Beckman-Coulter Super-speed ultracentrifuge Background: Essential hypertension (eHTN) affects millions of adults in the U.S. and contributes to end organ damage including kidney failure. The epidemic of eHTN is, in part, related to environmental changes, and in particular, the high sodium (Na) and cholesterol (chol) diets consumed in the U.S. The link between Na consumption and eHTN is established; however, the relationship between chol and eHTN is not well understood. Chronic, long-term hypercholesterolemia leads to arteriolosclerosis and diminished vascular compliance predisposing to eHTN; however, recent evidence suggests that acute, short-term chol ingestion augments renal Na avidity to raise blood pressure (BP). Hypercholesterolemia promotes incorporation of chol into lipid rafts (LRs) of the plasma membrane (PM) which raises PM viscosity and induces diseases such as diabetes (by suppressing insulin secretion) and diabetic nephropathy. The contribution of chol to organ dysfunction is underscored by the observation that chol chelation by cyclodextrin improved insulin secretion and diabetic nephropathy while treatment with statins had no effect. We hypothesize that chol incorporation into the lipid rafts and plasma membrane of the kidney collecting leads to enhanced epithelial sodium channel dependent transepithelial sodium transport which contributes to the development of hypertension. Objective: To isolate plasma membrane for isolation of lipid rafts and measurement plasma membrane cholesterol and plasma membrane protein expression, a high speed ultracentrifuge that generates up to 200,000g is required. Our research proposal requires the ability to isolate the plasma membrane from the other cellular components and to isolate lipid rafts from the plasma membrane. Travel: Not Applicable Special Material Requirements: Not Applicable Any Other Requirements unique to the solicitation: Not Applicable Scope of Tasks/Product Specifications: Our goal is to test whether cholesterol incorporation into the plasma membrane flow induced transepithelial Na transport in cortical collecting duct cells. This system will assist in the isolation of plasma membrane, and lipid rafts. Please see attached Document for brand justification. In particular the purchase of the Beckman Coulter model works with the currently available to us SW41 rotor (this is a Beckman Coulter product) that we have at the VA. Because we will get un updated rotor that can do larger volumes, the purchase of the Beckman Coulter will give our lab greater flexibility when performing our assays.
