- This is a Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to perform a potential requirement.This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition. It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract. Responses will not be considered as proposals or quotes. No award will be made as a result of this notice. The Government will NOT be responsible for any costs incurred by the respondents to this notice. This notice is strictly for research and information purposes only.Background: Cellular senescence is the reversible arrest of cell proliferation that happens when cells undergo stress potentially inducing abnormal proliferative events. Telomere shortening, genomic damage, and epigenomic damage, are all events that are associated with the induction of senescent phenotype. In parallel to arrest of cell proliferation, senescent cells are characterized by several changes in cellular metabolism and gene expression. Several proinflammatory cytokines, chemokines and growth factors are produced by the senescent cells; in general, they are defined as senescence-associated secretory phenotype (SASP), and they have both deleterious and beneficial effects, depending on the physiological context. SASP deleterious effects included induction of local inflammation; in contrast beneficial of SASP include stimulation of immune clearance and regulation of repair of damaged tissues.Human vascular smooth muscle cells (hVSMCs) undergo senescence when chronically exposed to cobalt chloride (a hypoxia mimic). In order to understand the mechanisms by which hVSMCs senesce, we want to measure the changes in both mRNAs and long non-coding RNAs comparing changes between treatment and over the duration of the exposure to cobalt chloride. In order to understand these temporal and gene expression changes, we plan to use a microarray to ensure we cover nearly all open reading frames as well as uncharacterized long non-coding RNAs.Purpose and Objectives: The purpose of the work requirement is to support projects regarding RNA regulation through lncRNA microarrays. The contractor will prepare sequencing libraries from the total RNA and use the prepared libraries for microarray analysis.The contractor will work closely with investigators and technicians to ensure that data are in the proper format and on the appropriate sequencing platform. The contractor will then provide FASTQ data files to the National Institute on Aging (NIA). Finally, the contractor will provide summaries and documentation to Government staff as necessary.Regular meetings between Government staff and the contractor will be scheduled either via teleconference or email. The contractor will be required to provide technical consultation and support as needed at the government staff meetings.Project requirements: The Contractor will perform the following:• RNA QC• cDNA synthesis• Target preparation by labeling with Cy3• Array hybridization, washing, and scanning• Data extraction, analysis and summarizationAnticipated period of performance: 1 year from date of awardCapability statement /information sought: Contractors that believe they possess the ability to provide the requirement described above should submit documentation of their ability to meet the project requirements to the Contract Specialist. The response should directly and specifically state, in the, capabilities statement, which project requirements you can supply. The capability statement should include 1) the total number of employees, 2) documentation of ability to provide the requirement, 3) any contractor GSA Schedule contracts by which all of the requirements may be met, if applicable, and 4) any other information considered relevant to this program.The respondent must also provide their DUNS number, organization name, address, point of contact, and size and type of business (e.g., 8(a), HubZone, etc., pursuant to the applicable NAICS code and any other information that may be helpful in developing or finalizing the acquisition requirements.One (1) copy of the response is required and must be in Microsoft Word or Adobe PDF format using 11-point or 12-point font, 8-1/2” x 11” paper size, with 1” top, bottom, left and right margins, and with single or double spacing.The information submitted must be in and outline format that addresses each of the elements of the project requirement and in the capability statement /information sought paragraphs stated herein. A cover page and an executive summary may be included but is not required.The response is limited to ten (10) page limit. The 10-page limit does not include the cover page, executive summary, or references, if requested.The response must include the respondents’ technical and administrative points of contact, including names, titles, addresses, telephone and fax numbers, and e-mail addresses.All responses to this notice must be submitted electronically to the Contract Specialist and Contracting Officer.
