Wide range of fluorescent filter capabilities to include the capability to capture: DAPI, GFP, Texas Red, Cy5, and TRITC wavelengths and standard bright field. Multiple objectives for a wide viewing range including 2X, 4X, 10X, 20X, and 40X, 60X (oil), and 100X (oil) magnifications, as well as 4X, 10X, 20X, and 40X phase contrasted ranges.Mounting adaptors to allow collection of images from slides, chambers, petri dishes, and tissue culture flasks. Custom modules to include Multi-stack, sectioning, image cytometer, time lapse.Navigation function where the observation point moves to a specific location on a wide-area map image, enabling high-magnification observation and an overall picture with a wide-area image keyed to the XY stage coordinate information. All mountable lenses shall allow the generation of a wide-area map image. Image stitching functionality (maximum of 50,000 x 50,000 pixels) captured via the automatic range set-up when using any mountable lens.Optical sectioning that is unaffected by fluorescence blurring during fluorescence imaging should be possible. Fully integrated microscope control software and image analysis for quantitation of captured still and video images. Software includes functionality that automatically selects cells based on contour and color contrast and automatically takes count and calculates the area, perimeter, Feret diameter, longest diameter, shortest diameter, and the integrated value of luminance. There should also be a function that uses the selected image information as a mask area to perform automatic counting upon re-setting the conditions for the images in each area, also enabling an easy measurement of area ratio, etc.The system should enable outputting, as a conditions file, of measurement conditions used with the automatic counting function. Furthermore, the system should have a macro processing function that performs automatic counting on multiple image files at once with the same conditions based on the conditions file.Equipped with a function that records videos at a frame rate of 95 fps or higher and images should be recorded at 12,000,000 pixels or more.Allows for mounting of temperature/CO2 regulated chamber for time-lapse imaging. The mounted chamber shall allow for multi-well plates to be set inside.Capable of transforming z-stacks into 3D renderings with a single click to accurately observe three-dimensional structures. Cytometer function that scans wells with uniform conditions for high reproducibility of data.Microscope, filters, and camera constructed inside a covered, Dark room simulated environmentThe features of the Axiovert A1 FL inverted microscope to include the following capabilities:Ability to visualize multiple muscle fiber types (up to 4 types simultaneously) using multi-channel fluorescent capabilities. Capture of muscle stem cell pre- and post-differentiated states, including size and cross-bridging analysis of mature myotubes. Capture of bone marrow stem cells.Imaging of mature osteoclast for quantitation and size measurement.Imaging of alizarin red and alkaline phosphatase stained osteoblast tissue culture plates.Imaging of oil-red-O staining of muscle fibers and liver histological samples for assessment of fat content.Capture of multi-tissue (Muscle, liver, adipocytes, cardiac, kidney) hemotoxilin and Eosin staining for structural and physiological assessment.Cell quantitation of seahorse extracellular flux analysis tissue culture plates to normalize cellular respiration data.Multi-color imaging of protein biomarkers in brain, muscle, liver, and bone tissues.Visualization of astrocytes and other brain derived cell types. Investigation of cellular RNA trafficking and looking into cells to investigate the kinetics and steady-state outcomes of catalytic RNA (ribozyme) therapeutics on specific target mRNAs and their cognate proteins.Investigation of real time ribozyme kinetics in live cells exploiting fluorescent RNA aptamer technology.Facilitate deeper investigations that include characterizing specific hematopoietic subtypes and macrophage polarization using the novel Z-stacking capabilities.Investigation of cardiac stem cell mediated repair using multiple fluorescence imaging and the quantitation of features to elucidate adult stem cells stimulation of endogenous myocyte proliferation.Imaging of iPSC-derived midbrain dopaminergic (DA) neurons, to better understand the impacts of parkin mutations as a model of idiopathic Parkinson s disease.Capture of high-resolution images of multi-channel fluorescent stains as well as automate imaging and quantification of large sections of tissue, thus augmenting the ability to compare the effectiveness of the exosome-based therapies as well as streamline the workflow process.The requirement will take place at a VA facility in Buffalo, NY. The sole source is pursuant to FAR 6.302-1 Only One Responsible Source and No Other Supplies or Services Will Satisfy Agency Requirements.
