(iii)�� �The solicitation document, the incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2020-06 effective 6 MAY 2020. (iv)�� �The associated NAICS code is 334516, Analytical Laboratory Instrument Manufacturing, and the small business size standard is 1,000. �The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13.5.� (v)�� �The Contractor Requirements are listed below: 1.0�� �PURCHASE ORDER TITLE Acquisition of a High throughput High Quality Direct Detection Gatan K3�� ��� � 2.0�� �BACKGROUND Cryo-Electron Microscopy (Cryo-EM) allows researchers to determine high-resolution 3D protein structures in a more native state (sub 3 Angstrom) without the need of crystallization. In recent years the field has witnessed a boom with its application to protein structure problems. The Multi-Institute cryoEM Facility (MICEF) currently possesses a high-resolution cryo-EM microscope, Titan KriosCryo Transmission Electron Microscope, that serves ~37 users from 18 labs in the four participating Institutes and Centers (ICs). �With this high demand each user will receive time on the Krios only once every 3 months, creating a large delay in data collection and productivity. �This severely hampered the research progress of the participating research programs and associated institutions.� possible laboratory based MX capabilities available as it allows development. 3.0�� �OBJECTIVE The scope of this purchase order is to purchase a K3 camera with BioQuantum Imaging capability and have it installed on the NIDDK�s existing Krios microscope. � The latest generation Direct Detection camera K3 from Gatan is significantly faster and has a larger imaging area than the current camera on the Krios (K2). �The K3 has a frame rate of 1500 frames per second (fps) compared to 400 fps for the earlier generation K2 camera, a 3.6X improvement in speed. The total pixels of the imaging area are 23.6 megapixels (5,760 x 4,092) compared to 14.4 megapixels (3,838 x 3,710) for K2, a 1.6X improvement in size. This results in a total of 6.2X improvement over the previous generation K2 camera. Therefore, upgrading the Krios to a K3 camera will more than triple the current output of the Krios and what is typically achieved in 3 days can be achieved in 1 day. This will greatly decrease the backlog of samples ready for data collection. 4.0 �� �SALIENT / REQUIRED FEATURES AND SPECIFICATIONS The K3 camera will replace the current K2 camera and will be mounted to the existing energy filter on the Krios microscope therefore, the successful offeror must provide a camera that is compatible with and will attach to �the Gatan energy filter on the existing Krios cyroEM microscope. The Gatan K3 attaches to the NIDDK�s Gatan energy filter on the existing Krios cyroEM microscope. �The readout speed of a Gatan K3 Direct Detection camera (1500 fps) surpass its competitors (all below 100 fps) by a large margin. This allows for both faster data collection and higher DQE data quality in Electron Counting Mode. Electron Counting Mode is well suited for Low Dose applications like Cryo EM for which the Cryo specimen (typically protein) is very sensitive to electron beam radiation damages. The specific items the NIDDK requires are listed below: K3 Camera/Installation: Part #�� ��� ��� ��� �Product Name 1967.U�� ��� ��� �BioQuantum K3 Imaging Filter Conversion Upgrade option that converts NIDDK�s existing Gatan GIF Quantum system to a K3-based energy-filtered direct detection system (963, 965, 967 upgrade to 1967) for high contrast imaging and tomography applications. Gatan installation is required. Completed to Gatan Inc.�s stated capabilities and performance specifications available at www.gatan.com/BioQuantumK3/datasheet. Includes one copy for online use, and one for offline use. (vi)�� �This requirement is being pursued through the authority granted in FAR 52.211-6, Brand Name or Equal.
