The section of Genetics of Organelle Biogenesis within the Laboratory of Biochemistry and Genetics/NIDDK is interested in understanding how cells control the number and size of organelles and focuses on centrioles as model to understand this problem.� Centrioles are small barrel-shaped organelles that are present as one or two pairs in each cell.� Our lab has been investigating how centriole size and number are controlled by mechanisms that operate at transcriptional and post-transcriptional levels to control the abundance of centriole components. Toward this end, we need to be able to accurately measure protein levels using quantitative immunoblotting which encompasses the use of fluorescently tagged antibodies and the accompanying instrumentation to image this fluorescence.� The iBright system will allow for the lab personnel within the NIDDK and quantitatively measure fluorescent immunoblots and thus is vital to our ability to successfully achieve our research goals� Responses to this notice shall contain enough information to establish the interested parties bona-fide capabilities for fulfilling the requirement and include certification by the OEM that you are authorized to provide services.� � �This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. �This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued.� � The solicitation number is 75N94020Q00012 and the solicitation is issued as a request for quote (RFQ). � The solicitation document, the incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2019-01 effective 20 FEB 2019. � �The associated NAICS code is 334516, Analytical Laboratory Instrument Manufacturing, and the small business size standard is 1,000 The acquisition is being conducted in accordance with the procedures of FAR Part 12 and FAR Part 13. � The Contractor Requirements are listed below: � Scope The iBright FL1500 imaging system capable of analyzing immunoblots stained with antibodies labeled with fluorophores in both the visible and infrared ranges, something that is not currently possible with existing instrumentation.� This system will allow us to measure multiple centriole components at once in a variety of different mutant backgrounds thus enabling us to efficiently interrogate the various mechanisms that ensure cells maintain a proper number of centrioles. The instrument must meet the following requirements in order to have sufficient capabilities to achieve the NIDDK research goals. Table-top imaging system with the following specifications: To simultaneously measure multiple centriole components, the system must be capable of imaging western blots in both the RGB and near infrared ranges and be able to image a 4-fold multiplexed western blot. Must be capable of imaging protein gels.� In order to detect low abundance centriole proteins, the system must be equipped with a highly sensitive, high resolution 9.1-megapixel CCD camera and have a wide range of binning options (1x1-8x8).� In order to ensure blots can be accurately quantified, the system should be equipped with an automated exposure tool to produce an image with the highest dynamic range. While minimizing pixel saturation.� To rapidly and carefully perform quantitative analysis of centriole proteins the system should be equipped with an image analysis software package and be coupled with highly automated onboard analysis support. Software should allow image contrast and background adjustment, background correction, direct lane and band identification, relative and absolute quantitation, as well normalization using housekeeping proteins or total lane protein measurements To image multiple blots at once, the imaging system should have a viewing area of at least 22.5 x 18.0 cm. � (viii)��� The provision at FAR clause 52.212-1, Instructions to Offerors � Commercial Items, applies to this acquisition. � (ix)���� The provision at FAR clause 52.212-2, Evaluation � Commercial Items, is applicable.� Award will be made to the contractor providing the most advantageous quote to the Government.� Technical factors and price considered.� The technical factor is as follows: The contractor will be evaluated on their product meeting the specifications as listed under the �scope�. � �(x)���� FAR clause 52.212-3, Offeror Representations and Certifications � Commercial Items, is applicable.� An offeror shall complete only paragraphs (b) of this provision if the offeror has completed the annual representations and certificates electronically via http://www.acquisition.gov.
