Page 2 of 2 Page 2 of 2 36C24221Q0578 SPECIAL NOTICE NOTICE OF INTEND TO SOLE SOURCE REAL TIME QPCR BUFFALO VAMC The Department of Veterans Affairs, Network Contracting Office NC02 in conjunction with the Buffalo VA Medical Center, 3495 Bailey Avenue, Buffalo, NY 14215, intends to solicit and negotiate with only one source under the authority of FAR 6.302-1(a)(2), Only one responsible source and no other supplies or services will satisfy agency requirements. The NAICS code is 334516 with a small business size standard of 1,000 employees. The VA intends to solicit and negotiate with Government Scientific Solutions, 12351 Sunrise Valley Drive, Reston, VA20191. This contract will be a Firm Fixed Price sole source for Real Time QPCR system. This system will be used to quantitatively measure hammerhead ribozyme (hhRz) cleavage reactions using optical fluorescence assays developed in the Sullivan lab. Steady state and single turnover reactions can be measured which combine cleavage and product leaving reactions. Binding reactions of ribozyme to substrate RNA will also be measured. Use of the Fluorescence Resonance Energy Transfer (FRET) capacity will also allow measures of conformational changes of the hhRz bound to substrate RNA which precede cleavage. Also, the instrument will be used to measure changes in gene expression in cells which are transfected with target RNA and hhRz to quantify the levels of target mRNA knockdown in comparison to catalytically inactive hhRzs or other control reactions We have identified a form of hammerhead ribozyme, which we call an Enhanced-hhRz (E-hhRz) which has greatly enhanced inherent kinetic speed than prior versions of this catalytic RNA. We are using catalytic RNAs as candidate RNA (or nucleic acid) therapeutics to suppress expression of target genes as a first step in developing therapeutics for diseases such as age-related macular degeneration (the focus of the VA Merit award). Up to this point all of our work has been done by manual pipetting of materials into cuvettes to initiate the reaction, which is measured by fluorescence optically in real time assays. This has opened a new technological space for RNA drug discovery. We anticipate now that even faster enzymes can be identified and optimized, and we need to test under a range of conditions that go beyond the physiological where the enzymes will function even faster. Moreover, we intend to apply our knowledge to new therapeutic disease targets to diversify this knowledge further. The qPCR machine will allow us to continue measurements of E-hhRz kinetics for various (diversified) targets which will extend our current knowledge to test if it is applicable to arbitrary target mRNAs that can occur in human diseases.
