The United States Department of Agriculture (USDA), Agricultural Research Service (ARS) is searching for firms who can provide a service contract labeled as follows: Statement of Work -Title: Metabolic Services Colorado State - Metabolomics Analysis As in past analyses, the Facility will perform the following: Liquid Chromatography Mass Spectroscopy. Inject 810 aqueous samples into a Waters Acquity UPLC CSH Phenyl Hexyl column (1.7 ?M, 1.0 x 100 mm) using a gradient from solvent A (2mM ammonium hydroxide, 0.1% formic acid) to solvent B (Acetonitrile, 0.1% formic acid) for separation. Gas Chromatography Mass Spec. 40 ?L aliquot of the aqueous phase re-suspension was dried under nitrogen, re-suspended in 50 ?L of pyridine containing 25 mg/mL of methoxyamine hydrochloride. Detect metabolites using a Trace 1310 GC coupled to a Thermo ISQ mass spectrometer (Thermo Scientific). Separate using a 30 m TG-5MS column (Thermo Scientific, 0.25 mm i.d., 0.25 ?m film thickness) with a 1.2 mL/min helium gas flow rate. Analyses and annotations. Define molecular features by retention time and mass-charge. Use the matchedFilter algorithm for GC-MS data, and the centWave algorithm for LC-MS data. Use RAMClustR to group features. Annotate GC-MS features by searching the GOLM metabolome database using the RAMSearch program. Annotate LC-MS features using RAMSearch and searches against the MassBank database using MSFinder.
