1 Statement of Work Multiclamp 700B Amplifier Systems 689-21-4-7155-0084 8/4/2021 Introduction/Overview This request is for two (2) Multiclamp 700B systems. The Multiclamp 700B amplifier is a computer- controlled amplifier capable of single-channel and whole-cell voltage patch-clamp and high-speed current clamp. It is an essential piece of equipment for experimental electrophysiology research laboratories. The research team will use this specialized equipment to perform essential electrophysiological experiments to identify neural circuits that are involved in the etiology of post- traumatic stress disorder (PTSD). Background Developing a better understanding and treatment for PTSD is a priority of the Veterans Health Administration (VHA). VHA has designated the National Center for PTSD (NC-PTSD), who partners with Yale School of Medicine, to accomplish this goal. Specific purpose research funding has been made available to the Clinical Neurosciences Division of the NC-PTSD, based at the VA CT Healthcare System during fiscal year 2021, to purchase specialized equipment which will better elucidate the neurobiological underpinnings of PTSD. As part of this work, several new research proposals are in development to link complementary technologies that study circuit function using animal models of PTSD in concert with neural circuit recordings and manipulation. By studying how different groups of neurons, whether intermixed in microcircuits or separated across brain regions, exert directional and causal influence on each other, mapping can be done to shape computations that underlie cognition and behavior. The MultiClamp 700B microelectrode amplifier is a current and voltage clamp amplifier for electrophysiology and electrochemistry research. This versatile amplifier is capable of single channel and whole-cell voltage patch-clamp, high-speed current-clamp (i.e. sharp electrode or field potentials), ion-selective electrode recording, amperometry/voltammetry, and bilayer recordings. The laboratory s current electrophysiology equipment for recording synaptic and circuit changes in brain tissue slices of PTSD mouse models, utilizes the older Axopatch 200A and 200B systems, which only allow a single channel recording and cannot easily switch between a voltage clamp and a current clamp. The Multiclamp 700B is by far the most widely used and trusted amplifier in the application of whole-cell slice recording. The equipment will be the property of the NC-PTSD/VA CT Healthcare System but will be located at the Yale Ribicoff laboratory and will be used by research investigators at each institution. Research protocols and projects that involve this equipment would require approval(s) from the Yale Institutional Animal Care and Use Committee (IACUC). Use of the equipment can commence shortly after installation and training, and we anticipate this will be a key resource for the psychiatric research community. Requirements The following are the minimum salient characteristics that must be met: Computer-controlled microelectrode current- and voltage-clamp amplifier designed to support up to two primary CV-7B headstages and two optional auxiliary (HS-2 or VG-2 type) headstages. Each CV-7B headstage contains a current-to-voltage converter for voltage-clamp and a voltage follower for true current-clamp. Ability to switch between low-noise patch-clamp recording and high-speed current-clamp recording. Compatible with Axopatch 200A and 200B system headstage setup. Compatible with CV-7B/BL headstage for bilayer recording. Features: External trigger command for programmable voltage threshold and rapid, automatic mode- switching between current and voltage-clamp. User-specified delay programming allowing for customization of recording procedures. Able to automatically detect current or voltage oscillations and disable or reduce compensation settings to protect the cell from damage. Able to automatically inject a compensatory current over a defined time course, in order to maintain cell membrane and avoid contamination of the clamp recording. Able to perform both voltage-clamp or current-clamp modes during the same experiment. CV-7B headstage with four different feedback resistors. In voltage-clamp mode, allows for a wide range of cellular recording. In current-clamp mode, includes three different current setting resistors to clamp current from a few pA up to 200 nA. Amplifier system can perform the function of two patch-clamp, two current-clamp, or a combination of patch- and current-clamp amplifiers. Dual headstages allow for complex synaptic experiments to be performed, as well as increasing throughput for drug discovery experiments, at a significantly lower cost per channel. Automation of pipette offset, fast/slow electrode capacitance compensation, whole-cell capacitance compensation, series resistance correction, pipette capacitance neutralization and bridge balance. Test values also can be directly entered into edit boxes, or selected from tables, depending on the field. Two-meter display output for voltage and current for each channel. Amplifier configurations can be saved and easily re-opened. MultiClamp Commander interface is not dependent on specific data acquisition software, and can be used with most data acquisition systems in standalone mode. Tasks Vendor should provide all new equipment free from damage and in working condition upon setup. This includes all items listed in the deliverables. Delivery The Multiclamp 700B system contains sensitive components and instrumentation. The additional transportation needed from the VA to Yale would increase the chance of damage during transportation, which would be costly and very time consuming. The request is that it be shipped directly to the site of research at the VA National PTSD Brain Bank/Ribicoff Lab, Department of Psychiatry at Connecticut Mental Health Center. It would be extremely difficult to inspect at the warehouse, repack for delivery to the research site, then schedule inspection again after delivery to the lab. Once delivered to the research site, the equipment will be inventoried and barcoded for annual scanning by VA personnel. Signed receiving paperwork will be released to the VA warehouse, along with serial number(s) for addition to the VA equipment inventory list.
