Description
This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6, as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation will not be issued. Solicitation number 12505B21Q0101 is issued as a request for quote (RFQ). The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular 2021-06. The associated NAICS code is 541380, Testing Laboratories, with a small business size standard of $16.5 Million and the PSC code is B529. The Department of Agriculture (ARS) has the need for the following services: 001) Hi-Fi PacBio DNA Sequencing of Alfalfa Genomic DNA Service Scope of Work: The USDA-ARS Plant Science Research Unit (PSRU) in St. Paul, MN are requesting quotes for Hi-Fi PacBio DNA sequencing of alfalfa genomic DNA. Background: � Developing genomic resources for alfalfa has lagged behind other crop species. There is currently no reference genome for alfalfa cultivars grown in the US. The lack of a reference genome sequence for tetraploid alfalfa impedes progress in molecular breeding methods to improve alfalfa including genomic selection, transgenic approaches to crop improvement, and genome editing. One goal of this project is to identify the core set of genes in alfalfa and the genes that are specific to plants with specific characteristics such as fall dormancy, winter survival and disease resistance. For this reason, we are interested in performing long read DNA sequencing from unique plants. Currently, several core facilities are providing this service and we would like to utilize one of these cores to perform long read DNA sequencing of alfalfa. The service provider will perform Hi-Fi PacBio DNA sequencing of alfalfa genomic DNA from plant samples provided by Plant Science Research Unit located at the University of Minnesota, St. Paul, MN. The goal of the ARS project is to identify the core and accessory genomes of alfalfa with emphasis on genes related to fall dormancy, winter survival, and disease resistance. Due to the tetraploid nature of alfalfa, each of the four sets of chromosomes has a different DNA sequence, requiring long reads to identify the four haplotypes. Technical Requirements:� Four (4) plant samples will be shipped to the service provider on dry ice. The service provider will perform these specific tasks: Isolate high molecular weight DNA from 4 plant samples as per industry standards for plant species. Perform qualitative analysis to ensure that DNA meets standard sequencing center QC standards for size, quality, and quantity. Prepare DNA libraries for sequencing from the above samples as per the manufacturer�s procedure for Hi-Fi DNA sequencing. Perform single-molecule real-time DNA sequencing using Pacific Biosciences SMRT cell technology on the Sequel IIe, with 30-hour Hi-Fi movies. Generate at least 64 Gb of Circular Consensus Sequence (CCS) DNA sequence data for each sample. Provide all raw and analyzed DNA sequence data for 4 plant samples via secure HTTP download. Delivery schedule: 90 calendar day from the time of contract award. The service provider is solely responsible for the entire service. There will be no support or use of federal facility by the service provider. The service provider will submit a report to end user at the end of the completion of the service. Data Rights:� All the data generated by the vendor belongs to the government. Evaluation Factors: Lowest price, technically acceptable. The government intends to award a firm-fixed price award based on the lowest price technically acceptable quote. The lowest priced quote will be evaluated first; if not found acceptable, the next lowest priced quote will be evaluated, and so on until an acceptable quote is identified. The government will not review higher priced quotes if the lowest priced quote is determined to be technically acceptable. For a vendor�s quoted products to be considered technically acceptable, they must meet all of the technical requirements identified in the solicitation/SOW. Key Deliverables: Isolate High Molecular Weight DNA Qualitative Analysis DNA Libraries for Sequencing Single-Molecule Real-Time DNA Sequencing 64 Gb of Circular Consensus Sequence (CCS) DNA Sequence Data All Raw and Analyzed DNA Sequence Data Delivery: All data generated by the vendor will be submitted to Deborah Samac at debby.samac@usda.gov within 90 calendar days from date of contract award.� The Government anticipates award of a Firm Fixed Price contract. Submitting a Quote: To be eligible for an award, all contractors must be registered in the System for Award Management (SAM) with a current and active registration. A contractor can contact SAM by calling 1-866-606-8220 or e-mail at www.sam.gov. NO EXCEPTIONS. A DUNS (Dun and Bradstreet) number is required in order to register.�Vendor must also include their DUNS# on their quote. Finally, to be eligible, all vendors must be NDAA 889 Certified. If they do not currently have a NDAA 889 Certification, then they must complete the attached NDAA 889 Certification form and submit it to aaron.dimeo@usda.gov so that they can be added to the list of NDAA 889 Certified vendors. The submission and payment of invoices on the resulting contract will be made through the Invoice Processing Platform (IPP). As such, vendors must be currently registered or register in IPP in order to be able to submit and receive payment on invoices for the resulting contract. � Offerors responding to this announcement shall submit their quote on�SF 1449�(see attached). The quote should include all fees, delivery charges, and/or surcharges. This will be a tax-exempt purchase. Only electronic submissions will be accepted. Please email all quotes in writing on the SF 1449 to�aaron.dimeo@usda.gov by no later than�Thursday, August 19, 2021 at 5:00 PM�Central Standard Time. Questions in regards to this combined synopsis/solicitation must be submitted through email and are due no later than�5:00 PM CST on Friday,�August 13, 2021.
