- This notice is being published in accordance with Federal Acquisition Regulation (FAR) Part 5.101(a)(2) requiring the dissemination of information on proposed contract actions expected to exceed $15,000, but not expected to exceed $25,000. This is a notice of intent to award a sole source contract to Synthego�and is not a request for competitive proposals.� This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures, Subpart 13.106-1 (b) (1), Soliciting from a single source and is not expected to exceed the simplified acquisition threshold.� Contracts awarded using FAR Part 13�Simplified Acquisition Procedures and at a dollar value less than the simplified acquisition threshold are exempt from the requirements of FAR Part 6�Competition Requirements. Single-sole Source Determination: The determination by the Government to make a single-sole source�is based upon the market research conducted as prescribed in FAR Part 10�Market Research, specifically the results of the market research concluded that this order is for knockout single cell clone�offered by Synthego.� To better understand how alterations in glycosylation contribute to disease onset and progression, the Developmental Glycobiology Section studies how sugar addition (O-glycosylation) is regulated and how it influences basic biological processes. In this project, we want to knockout single O-glycosyltransferase in specific cell lines to find which proteins are modified by O-glycosylation and understand the biological function of this modification. The ability to find the proteins with O-glycosylation modification will greatly increase our understanding of their function during development in vivo. Additionally, these knockout cell lines will highly beneficial for biological function analysis of O-glycosylation. Synthegos company is the only company that exclusively utilize sgRNA and Cas9 protein to form an RNP to perform editing, which leads to higher rates of success, shorter timeframes, and lower cost. To design the sgRNA, a crucial component of the process, the company utilizes a proprietary and purpose-built multiplexing strategy that no other company has access to. The multiplex format uses up to three chemically modified sgRNAs that are designed to cause at least one fragment deletion (21+ bp). � Contractor requirements: For making O-glycosyltransferase knock out cell lines, detailed steps for each gene knock out cell line making include: 1. Design and Synthesis of modified sgRNA and donor template (for knock-in projects). Contractor�will design several guides targeting the region of interest. Each guide will be required to meet strict off-target requirements of at least two mismatches, within an early exon and targeting a common exon present in the majority of annotated transcripts. Guides will be synthesized onsite by contractor�as chemically modified synthetic single-guide RNA. 2. Clonal isolation and initial genotyping. The edited pool is used to seed single cells for clonal expansion. Each well seeded is imaged every 2-3 days and rigorously tracked to ensure the population is truly clonal and only the progeny of a single cell. The resulting clones are verified using Sanger sequencing. 3. Sequence validation of alleles Colonies identified in Step 2 will be further screened and sequenced to precisely identify those that harbor the edits required. Two clones will be selected to be expanded for final QC. The Quality Control process confirms genotype via Sanger sequencing and confirms the absence of mycoplasma. Closing Statement: This notice is not a request for competitive proposals.� However, interested parties may identify their interest and capability to respond to this notice.� Responses to this notice shall contain sufficient information to establish the interested parties� bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size.
