- This notice is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Federal Acquisition Regulation (FAR) Subpart 12.6 �Streamlined Procedures for Evaluation and Solicitation for Commercial Items,� as applicable, and as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; quotes are being requested and a written solicitation document will not be issued. Submit offers on RFQ-RML-C-2088052 � �This solicitation is a request for quotes (RFQ). The solicitation documents and incorporated provisions and � � � �clauses are those in effect through Federal Acquisition Circular FAC 2021-06 Effective July 12, 2021 � �The associated North American Industry Classification System (NAICS) code for this procurement is 325414 with a small business size standard of 1250 employees. This acquisition is being posted as a Brand Name Restriction with �No� small business set-aside. The National Institute of Allergy and Infectious Diseases (NIAID), The Laboratory of Virology at Rocky Mountain Laboratories is seeking to purchase Immunogenomics 10x B-cell Single Cell Immuno-Profiling and related services. The Viral Pathogenesis Section (VPS) routinely surveys the dynamics of B-cell responses following Zika virus (or other heterologous flavivirus) infection or vaccination. We need efficient, accurate, sensitive and prompt ultra-deep sequencing and customized bioinformatics to provide comprehensive characterization of the immense diversity of B-cell receptors (BCR) repertoire, including the ability to pair heavy and light BCR chains at single-cell resolution. Genewiz 10X B-cell Single Cell Immuno-Profiling and related services includes single cell isolation via microfluidic technology with up to 65% cell capture efficiency and subsequent indexed sequencing on Illumina Sequencing 10X Configuration. Sequencing would include the processing of about 6,000 cells per sample, with 5,000 total number of reads per cell. They also offer a proprietary cell freezing protocol that maintains cell viability and the company also performs a dead cell removal process that improves data quality. Service includes Ph.D.-level consultation providing guidance before, during and after the project, as well as the automated mining of data to yield easy-to-interpret results that reduces analysis time for the researcher. Additionally, large data files are stored for up to six months and can easily be accessed and analyzed by 10x Genomics Cell Ranger and Loupe Cell Browser allowing for customization of the sequencing results and easy sequence transfer for downstream DNA gene synthesis, if needed. Additionally, the service can be paired with other Genewiz services such as Bulk Immuno-profiling and downstream DNA gene synthesis that streamlines data processing and analysis from a single study. Statement of Work This statement of work is for the procurement of Immunogenomics 10x B-cell Single Cell Immuno-Profiling and related services. The Viral Pathogenesis Section (VPS) routinely surveys the dynamics of B-cell responses following Zika virus (or other heterologous flavivirus) infection or vaccination. We need efficient, accurate, sensitive and prompt ultra-deep sequencing and customized bioinformatics to provide comprehensive characterization of the immense diversity of B-cell receptors (BCR) repertoire, including the ability to pair heavy and light BCR chains at single-cell resolution. Our specific requirements are enumerated below: 1.�� �Government shall provide at least 50,000 cells in 0.5 mL aliquot per sample, submitted in duplicate as frozen single cell suspension with at least 90% cellular viability after thawing. 2.�� �Vendor shall provide cryopreservation protocol to aid shipment of samples. 3.�� �Vendor shall be able to perform single cell isolation via microfluidic technology with up to 65% cell capture efficiency. 4.�� �Vendor must perform initial sample QC free of charge, including the following: a.�� �Assessing RNA integrity by Tapestation and concentration by Qubit assay� b.�� �Assessing library size on the Agilent Tapestation, concentration by Qubit assay, and final quantitation by qPCR. 5.�� �Vendor shall perform indexed sequencing on Illumina Sequencing 10x Configuration targeting BCR genes in pre-defined primer sets. 6.�� �Vendor shall perform sequencing in about 6,000 cells per sample. 7.�� �Sequencing yield shall be about 5,000 total number of reads per cell. 8.�� �Multiplexing of samples must be performed such that samples are run on the same chip whenever possible. 9.�� �Vendor shall supply quality reports, FASTQ files and bioinformatic tools, including interactive V(D)J analysis through the 10x Genomics Cell Ranger and Loupe Cell Browser. 10.�� �Vendor must provide a dedicated PhD-level Study Manager and weekly project updates. 11.�� �Vendor must offer sample storage for at least one year following project completion. 12.�� �Vendor must offer data storage of FASTQ files for up to 6 months. 13.�� �data files shall be stored for six months and must be accessed/analyzed easily by 10x Genomics Cell Ranger and Loupe Cell Browser. Allowing for customization of the sequencing results and easy sequence transfer for downstream DNA gene synthesis, if needed. 14.�� �This service shall be capable of being paired with other Genewiz services. For Example, Bulk Immuno-profiling and downstream DNA gene synthesis that streamlines data processing and analysis from a single study. NIAID will make that determination based on the capability of each vendor�s quote to provide all requested products/services, meet the brand name requirements, provide the proposed products/service in a timely manner, warranty, and reasonable price. The Government�s objective is to obtain the highest technical solution considered necessary to achieve the project objectives with a reasonable price.
