Description
**AMENDMENT TO EXTEND SOLICITATION DATE ** This is a combined synopsis/solicitation for services in which the government intends to acquire in accordance with FAR Part 13, Simplified Acquisitions Procedures. Department of Commerce (DOC), National Oceanic & Atmospheric Administration (NOAA), National Marine Fisheries Service (NMFS), Alaska Fisheries Science Center, Auke Bay Laboratories. Researchers at the Alaska Fisheries Science Center use DNA sequencing in a variety of ways to better understand and manage fish stocks. DNA sequencing will be used for continuing assessment of genetic population structure of important assessed species to inform stock assessment. To conduct this sequencing, we need high-quality, chromosome level genome assemblies for species of interest. We require an initial high-quality genome assembly derived from a tissue sample then additional sequencing (Hi-C or similar) to achieve complete and chromosome level assemblies. The services we require include library preparation (including quality control), deep sequencing of prepared libraries, chromosome level genome assembly, phasing of reads, and bioinformatics for assembly into chromosomes. The two species we require chromosome level assemblies for are sablefish (Anoplopoma fimbria) and snow crab (Chionoecetes opilio). For the two species noted above, we require 1) assembly of draft genomes from tissue samples including library preparation, sequencing with long-read technology, and bioinformatics to construct an initial assembly that can be anchored to chromosomes, 2) sequencing of Hi-C or similar libraries including library preparation using a workflow such as proximo sequencing or similar, (3) phasing of reads using the FALCON-phase workflow or similar 4) bioinformatics to merge the initial draft genome with Hi-C or similar sequencing to construct high-quality chromosome level assemblies. Sequencing cannot be completed in house because we do not have the equipment and it is prohibitively expensive (millions of dollars). We are therefore setting up a contract to receive genome sequencing and assembly as a service. Our goal is to obtain high-quality reference genomes for important assessed species to inform management of these species. In other words, we are leveraging sequence data to investigate stock structure of important species, which will provide information to aid in their management. A critical component of the NMFS mission is to aid in the management of commercially harvested fish and this sequencing contract will help us accomplish this mission. �See attached Statement of Work for more information.� �All interested concerns must submit a quotation and documentation demonstrating the qualifications to provide services as described above within fifteen (15) days of the publication date of this notice. This announcement constitutes the only solicitation, quotations are being requested and a written solicitation has been issued. A firm-fixed order is anticipated to be issued in within 24 hours of closing date and time.
