SOURCES SOUGHT NOTICE Solicitation Number: 75N95022Q00109 Title:� A microscope system with Large Field of Viewpoint scanning confocal and widefield video recordings and spatial optogenetics stimulation Classification Code:� 6640 - LABORATORY EQUIPMENT AND SUPPLIES NAICS Code:� 333314 - Optical Instrument and Lens Manufacturing Size Standard: 500 Employees Description:� The National Center for Advancing Translational Sciences (NCATS), Division of Pre-Clinical Innovation (DPI) is in need of a microscope system with Large Field of Viewpoint scanning confocal and widefield video recordings and spatial optogenetics stimulation to implement drug screening assays on engineered tissue models of the neuromuscular junction using the CuriBio Mantarray muscle contraction platform and of neuronal calcium activity on neural circuit models. This is a Small Business Sources Sought notice. This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding: (1) the availability and capability of qualified small business sources; (2) whether they are small businesses; HUBZone small businesses; service-disabled, veteran-owned small businesses; 8(a) small businesses; veteran-owned small businesses; woman-owned small businesses; or small disadvantaged businesses; and (3) their size classification relative to the North American Industry Classification System (NAICS) code for the proposed acquisition. Responses must include: (1) the place of manufacturing (i.e. address if supply/equipment is a domestic end product and include country of manufacture), as well as (2) the size status of the manufacturer under the applicable NAICS code (i.e. address Non-Manufacturer Rule). Your responses to the information requested will assist the Government in determining the appropriate acquisition method, including whether a set-aside is possible. An organization that is not considered a small business under the applicable NAICS code should not submit a response to this notice. This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition.� It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract.� Responses will not be considered as proposals or quotes.� No award will be made as a result of this notice.� The Government will NOT be responsible for any costs incurred by the respondents to this notice.� This notice is strictly for research and information purposes only. Background:� The National Institutes of Health (NIH) is the nation�s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and apply that knowledge toward making and supporting medical discoveries that improve people�s health, reduce illness and disability, and lengthen and save lives. The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), which mission it is to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions. The 3D Tissue Bioprinting Laboratory at NCATS is producing and using engineered tissues as physiological relevant assay platforms for predictive drugs screening. This is a request for a new microscope which will be applied to develop functional neuronal tissue models, including neuronal circuits of addiction and innervated skin models (HEAL program), neuromuscular junction (NMJ) tissue and related diseases (potential application for gene therapy, like Congenital myasthenic syndrome (CMS) for PaVe-GT program), analyze the contractibility of the NMJ Model, contractibility in cardio muscle tissue models for toxicity studies (in collaboration with Tox21). As well as being the labs primary equipment to develop protocols for optogenetic analysis and then translating to high-throughput equipment & several robotic systems that NCATS has. With these new capabilities and the current instrumentation such as Phenix Plus, Molecular Devices ImageXpress and Leica confocal do not allow to implement such measurements, because of limitations in hardware and software. As well as camera quality & speed and also because the commercial platform used to develop the NMJ (CuriBio) is too tall (29mm) and does not physically fit any other imaging instrument at NCATS.� Product Description: The requirements for the microscope system with Large Field of Viewpoint scanning confocal and widefield video recordings and spatial optogenetics stimulation shall be Brand Name or Equal. Please refer to attachment. Project requirements: At a minimum, the salient physical, functional, or performance characteristics that equal� products must meet are as follows: This microscope must be able to: Do both fluorescence confocal and widefield, while also maintaining a large field of view (FOV) brightfield video image collection, as well as spatial optogenetics stimulation capabilities to measure light-stimulated muscle contraction and neural activity on bioengineered muscle and neural issue models, respectively. The microscope must be compatible and have room in the sample holder to physically fit the custom CuriBio Mantarray pillar muscle contraction plate currently used at NCATS (e.g. the Nikon TI2-E inverted microscope is already being used by CuriBio and we know is compatible with the Mantarray plate); the largest FOV of at least 25mm FOV (e.g. Nikon Ti2-E microscope has a 25 mm FOV from the sideport of the microscope) to be able to measure contraction from the whole muscle tissue in the Mantarray plate; and have a camera compatible with the larger FOV and higher pixel resolution (e.g. Photometrics Kinetix sCMOS, 29.4mm FOV) that enables brightfield video records of whole muscle tissue contraction. With the muscle tissue being suspended 2.5mm inside the well, this requires a large FOV microscope. This microscope must be able to provide fast 3D tissue imaging with point-scanning confocal microscope with higher resolution and increased speed, larger field of view (25mm), with scanning sizes up to 8192 x 8192 pixels, with 2X objectives, and capability for supervideo frame rate (high-speed imaging with up to 720 fps) to measure calcium transient in both muscle and neural tissue models. Speed is important for the calcium transient and contractibility measurements in the NMJ project, which requires a higher fps camera to accurately capture and measure these results. The microscope must be able to do spatially targeted photo stimulation with a digital mirror device (DMD). 5 channel wavelengths, multi-region illumination, all simultaneously, and at a subcellular resolution. Can be directly applied to the NMJ tissue model for stimulation of the neuron-only while simultaneously recording the innervated muscles contraction or engineered brain-region specific neural circuits. The device must allow for the advancements in microfabrication with capabilities in photopatterning hydrogels utilized in many projects. With the broad wavelengths available, this will allow for multi material constructs in the models such as NMJ and the Innervated Skin project. The microscope must have a motorized stage for automated measurements from a multiwell plate, and a customizable plate library for unique and custom labware. The microscope must have an enclosure with humidity, temperature and CO2 control. Anticipated period of performance: On or before April 30, 2022. Other important considerations: Cite any other information that is necessary for potential respondents to understand the nature of the potential requirement or proposed acquisition. Capability statement /information sought. Small business concerns that believe they possess the capabilities to provide the required products or services should submit documentation of their ability to meet each of the project requirements to the Contracting Officer. The capability statement must specifically address each of the project requirements separately.� Additionally, the capability statement should include 1) the total number of employees, 2) the professional qualifications of personnel as it relates to the requirements outlined, 3) any contractor GSA Schedule contracts and/or other government-wide acquisition contracts (GWACs) by which all of the requirements may be met, if applicable, and 4) any other information considered relevant to this program. Small businesses must also provide their Company Name, DUNS number, Physical Address, and Point of Contact Information. Interested small businesses are required to identify their type of business, applicable North American Industry Classification System (NAICS) Code, and size standards in accordance with the Small Business Administration. The government requests that no proprietary or confidential business data be submitted in a response to this notice. However, responses that indicate the information therein is proprietary will be properly safeguarded for Government use only. Capability statements must include the name and telephone number of a point of contact having authority and knowledge to discuss responses with Government representatives. Capability statements in response to this market survey that do not provide sufficient information for evaluation will be considered non-responsive. When submitting this information, please reference the solicitation notice number. The respondent must also provide their DUNS number, organization name, address, point of contact, and size and type of business (e.g., 8(a), HubZone, etc., pursuant to the applicable NAICS code and any other information that may be helpful in developing or finalizing the acquisition requirements. One (1) copy of the response is required and must be in Microsoft Word or Adobe PDF format using 11-point or 12-point font, 8-1/2� x 11� paper size, with 1� top, bottom, left and right margins, and with single or double spacing. The information submitted must be in and outline format that addresses each of the elements of the project requirement and in the capability statement /information sought paragraphs stated herein.� A cover page and an executive summary may be included but is not required. The response is limited to ten (10) page limit.� The 10-page limit does not include the cover page, executive summary, or references, if requested. The response must include the respondents� technical and administrative points of contact, including names, titles, addresses, telephone and fax numbers, and e-mail addresses. All responses to this notice must be submitted electronically to the Contract Specialist and Contracting Officer.� Facsimile responses are NOT accepted.
