- This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. The solicitation number is HHS-NIH-NIDA-CSS-75N95022R00045 and the solicitation is issued as a request for proposal (RFP). This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures; �and FAR Part 12�Acquisition of Commercial Items, and is not expected to exceed the simplified acquisition threshold. The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) Number 2022-04, with effective date 1/30/2022. The associated NAICS code is 325414 and the small business size standard is 1,250 employees. This requirement is full and open with no set-aside restrictions. This requirement is for the following: Statement of Need and Purpose: The purpose of this requirement is to design a CRISPRi sgRNA library that improves upon the current research protocols, resulting in more robust gene knockdown and allowing more compact screens, thus reducing the cost of future experimentations. Background Information and Objective: The objective of this requirement is to generate a pooled oligo library of sgRNAs for the purpose of performing CRISPRi survival screens in iPSC differentiated into neurons for the Neurodegenerative Initiative (iNDI) project. The iNDI project has engineered a set of iPSC lines. Using these engineered lines, the generation of this tool allows for a CRISPRi sgRNA library to improve upon the current research protocols. This will result in a more robust gene knockdown and allow more compact screens.� This entails a multi-step cloning method performed at a large-scale to ensure that the final library contains all of the ~20,000 unique sgRNAs that are included within the oligo pools. This requires a high level of expertise and quality control. Project requirements: Requirement#1 Generate a cloned oligo pool library of iC20-dsgRNA-2xU6-UMI-bsd-BFP_1, 21554 customer-defined variants cloned into customer-supplied vector, 250 ug plasmid DNA to be delivered to the INDI researchers. The synthetic oligo production needs to be the same in form fit and function of existing oligos used in this research project to maintain continuity. Requirement#2 Generate a cloned oligo pool library, iC20-dsgRNA-2xU6-UMI-bsd-BFP_2, assembly of 13569 UMIs, and constant region into customer-supplied vector with custom quality control, to be delivered to the INDI researchers. The synthetic oligo production needs to be the same in form fit and function of existing oligos used in this research project to maintain continuity. The Government anticipates award of a firm fixed-price contract for this acquisition, and the anticipated period of performance is one month after award � Delivery or Deliverables: For each of the of the supplied custom vector, vendor will produce 2 oligo pools creating one final library containing all of the guide RNAs and UMIs;� ~20,000 unique guide RNAs and 13, 000 UMI�s that are being currently researched within the CARD initiative. Key Personnel: Vendor will provide a dedicated project manager to the government as a point of contact for the services. Technical and past performance, when combined, are significantly more important than cost or price. A written notice of award or acceptance of an offer, mailed or otherwise furnished to the successful offeror within the time for acceptance specified in the offer, shall result in a binding contract without further action by either party. Before the offer�s specified expiration time, the Government may accept an offer (or part of an offer), whether or not there are negotiations after its receipt, unless a written notice of withdrawal is received before award. Responses to this solicitation must include clear and convincing evidence of the offeror�s capability of fulfilling the requirement as it relates to the technical capability The price proposal must include unit price, shipping and handling costs, delivery days after contract award, delivery terms, breakdown and rationale for proposed costs, and any other information or factors that may be considered in the award decision.
