This is NOT a solicitation for proposals, proposal abstracts, or quotations. The purpose of this notice is to obtain information regarding the availability and capability of all qualified sources to perform a potential requirement. This notice is issued to help determine the availability of qualified companies technically capable of meeting the Government requirement and to determine the method of acquisition. �It is not to be construed as a commitment by the Government to issue a solicitation or ultimately award a contract. �Responses will not be considered as proposals or quotes. �No award will be made as a result of this notice. �The Government will NOT be responsible for any costs incurred by the respondents to this notice. �This notice is strictly for research and information purposes only. Background: �NCATS has become a key partner in the Antiviral Program for Pandemics (APP), a multi-agency initiative that includes NIAID and BARDA, to accelerate antiviral development through early discovery and preclinical development of safe and effective oral antivirals. The initial priority for the APP is to develop treatments for SARS-CoV-2 and other coronaviruses, with the program expanding to address other virus families with pandemic potential. NCATS will apply its proficiency in drug discovery and development and its cutting-edge technologies to fill critical needs of the APP, such as (i) target validation, (ii) high-throughput screening (HTS) for drug starting points, (iii) medicinal chemistry for lead optimization, and (iv) preclinical, investigational new drug (IND)-enabling development. APP chemists within DPI are tasked with the discovery and development potential small molecule antiviral treatments for a range of viruses that have pandemic potential. In order to meet the increased demand for sample purification and analysis, the Analytical Chemistry Core (ACC) needs to improve throughput and expand capabilities through procurement of additional instrumentation and equipment, as well as the application of new technologies. Purpose and Objectives: The concepts of throughput and cycle time are of great significance to translational scientists as they endeavor to increase experimental capacity while reducing the therapeutic discovery and development timeline in an effort to rapidly bring more treatments to patients. The development of an efficient method for the transfer of compounds from Medicinal Chemistry to Compound Management (CoMa) enables a faster turnaround between small molecule synthesis and biological assays. The compound workflow consists of 5 stages (synthesis, purification, processing, registration, and storage) each comprised of multiple steps. The ACC is responsible for purification, processing, and registration and has created the SMART Centralized Sample Purification and Processing Platform where internally synthesized compounds progress from sample submission to CoMa in only 5 days (2 days for purification, 2 days for processing, and 1 day for registration). The rationale is that by having the ACC conduct the majority of sample purifications the medicinal chemists are afforded more time to focus on synthesizing compounds and thus establishing a streamlined compound synthesis workflow, which is more efficient and productive. Column chromatography is a single-sample-at-a-time process often making purification a major bottleneck in the medicinal chemistry pipeline. Despite this fixed parameter, advancements can be made with instrumentation, automation, methods development, and systems integration to create the most efficient purification workflow possible. The ACC has designed the FluidCore automated purification system as our solution for the technological problem of sample throughput. The ACC worked with Tecan on the design and development of an innovative and custom sample management and fraction collection platform built around the Tecan Freedom EVO150 liquid handling system. This custom Tecan Freedom EVO150 utilizes a dual arm fraction collection bed capable of accommodating a wide range of sample vessels, which can be arranged in a variety of configurations. The FluidCore system consists of the custom Tecan Freedom EVO150 platform connected with Waters analytical and purification LC instrumentation creating a single platform for automated purification. By developing the FluidCore automated purification system, current manual steps of the workflow will be replaced with automated protocols. The integrated Waters analytical UPLC-MS will be used to analyze pre-purification samples for automated method development. The Waters preparative LCMS will then use this method for purification of the sample where individual fractions are dispensed into containers on the custom Tecan Freedom EVO150 fraction collection bed followed by automated fraction analysis by the analytical UPLC-MS. Performance of all these processes within the same platform in an automated fashion will improve operational efficiency and reduce the cycle time for a compound to go from synthesis to plating in an effort to accelerate the drug discovery process. The custom Tecan Freedom EVO150 liquid handling platform is required as it is the core component of the ACC FluidCore automated purification system.
