PRE-SOLICITATION NOTICE - NOTICE OF INTENT NOI-NIAID-2132046 NAICS � 541380 � Testing Laboratories This is a Notice of Intent, not a request for proposal. The National Institute of Allergy and Infectious Diseases (NIAID) of the National Institutes of Health (NIH) intends to negotiate on an other than full and open competition basis with Applied Stemcell, Inc. of Milpatis, CA to generate two point mutation cell lines using the CRISPR/Cas9 platform. The final deliverable will be two (2) clones that are homozygous for the targeted allele, with two (2) vials of cells at 1M cells/vial, and a final report containing details of targeting design, experimental and genotyping results. Laboratory of Persistent Viral Disease (LPVD) studies the role of persistent infection in the development of immunosuppression and neurological disease. The prion cell biology unit uses a polymorphism at amino acid 129 in the prion gene influences the disease subtype of an infecting prion.� For the investigation of different prion disease subtypes, cells in a match clonal background are required.� The provider must use supplied heterozygous iPSCs for cloning, and generate two homozygous clones of the 129MM and 129VV genotypes (two vials of each clone, 1 x 10^6 cells/vial) and two isogenic clones without the mutation (two vials of each clone, 1 x 10^6 cells/vial).� Pluripotency and gene sequence of cells must be validated accordingly. This is a follow-on requirement to procurement additional quantities of previously generated cell lines. The statutory authority for this sole source requirement is 41 U.S.C. 253 (c) (1) as implemented by FAR 13.106-1 only one responsible source and no other supply of service will satisfy agency requirements. THIS IS NOT A REQUEST FOR PROPOSAL.� All responsible sources that could provide comparable services may submit a capability statement that will be considered by email (subject line to reference NOI-NIAID-2132046) to Dana Monroe at dana.monroe@nih.gov by 5:00 pm MDT Tuesday, July 5, 2022.� All responses received by the closing date of this synopsis will be considered by the Government.� A determination not to compete this requirement, based upon responses to this notice, is solely within the discretion of the Government.
