- NOTICE OF INTENT to Sole Source SOLICITATION NUMBER: NIMH-005125-NOI TITLE: NIMH 10X Genomics Library Prep Kits CLASSIFICATION CODE:65 Medical/dental/veterinary Equipment/supplies NAICS CODE: 334516 Analytical Laboratory Instrument Manufacturing RESPONSE DATE: Due 7/28/22 PRIMARY POINTS OF CONTACT: Kyle Miller Kyle.miller2@nih.gov Christine Frate Christine.frate@nih.gov DESCRIPTION: INTRODUCTION THIS IS A PRE-SOLICITATION NON-COMPETITIVE (NOTICE OF INTENT) SYNOPSIS TO AWARD A CONTRACT WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME). The National Institute of Mental Health (NIMH) Human Genetics Branch (HGB) intends to negotiate on a sole source basis with 10X GENOMICS INC for the purchase of library prep kits in order to prepare single-cell sequencing libraries for 80 samples. NORTH AMERICAN INDUSTRY CLASSIFICATION SYSTEM (NAICS) CODE and SET ASIDE STATUS The intended procurement is classified under NAICS code 334516 Analytical Laboratory Instrument Manufacturing with a Size Standard of 1,000 employees. REGULATORY AUTHORITY This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13Simplified Acquisition Procedures. Contracts awarded using FAR Part 13Simplified Acquisition Procedures are exempt from the requirements of FAR Part 6Competition Requirements. FAR Subpart 13.106-1 (b) Soliciting from a single source provides that: For purchases not exceeding the simplified acquisition threshold, Contracting officers may solicit from one source IF the contracting officer determines that the circumstances of the contract action deem only one source reasonably available (e.g., urgency, exclusive licensing agreements, brand-name OR industrial mobilization). The resultant contract will include all applicable provisions and clauses in effect through the Federal Acquisition Circular (FAC) 2022-06 dated April 26, 2022. DESCRIPTION OF REQUIREMENT The National Institute of Mental Health (NIMH) Human Genetics Branch (HGB) are requesting the library prep kits in order to prepare single-cell sequencing libraries for for 80 samples, 64 as single-cell RNA-seq and 16 as a combination of single-cell RNA-seq and ATAC-seq. The samples will be processed by the National Human Genome Research Institute (NHGRI) Microarray and transcriptomics Core. A major focus of our laboratory is to study the impact of genetic risk alleles on gene expression in postmortem human brain and in neural cells derived from induced pluripotent stem cells. Toward this end, we have previously spent several years studying the effect of psychiatric risk allele in bulk RNA sequencing. However, the insights we could obtain in bulk tissue are limited by its inherent heterogeneity and averaging across variable mixtures of cell types. These severe shortcomings of bulk RNA sequencing can now be overcome using single-cell RNA-sequencing. By sequencing individual cell populations in genetic risk allele carriers, we will be able to resolve cell types and study the effect of genetic risk alleles on specific cell populations in post-mortem human brain and different stages of neuronal differentiation in our iPSC-derived models and brain samples from animal models. To study potential transcriptional mechanisms underlying the gene expression changes, we will use a combination of gene expression and open chromatin profiling for the profiling of single cells in the postmortem brain samples. This project will last one (1) year after first delivery with the product staggered until on or before July 31, 2023 so as to ensure prompt use of perishable reagents. CONTRACTING WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME) DETERMINATION During market research, it was found that 10x Genomics library prep kits is the unique compatibility with equipment present at NHGRI Microarray and transcriptomics Core and comparability to other single-cell data previously obtained in the lab and at the Human Brain Collection Core (HBCC) using the same system. The 10x Genomics platform is the only available tool for completing the desired type of single-nucleus sequencing. There is no other vendor than 10x Genomics that can provide the required library kits. The materials are needed for a single-cell sequencing project that can only be run using the 10x system hence the need to purchase materials from 10x Genomics. The equipment available to us to perform this experiment on a large scale and with contained costs of reagents and personnel time is a 10X Genomics microfluidics system at the Microarray and transcriptomics Core. Therefore, with these facts in mind, it was found that 10x Genomics is the only vendor that can provide the needed products. The intended source is: 10x Genomics, Inc. 6230 Stoneridge Mall Road Pleasanton, CA 94588-3260 CLOSING STATEMENT THIS SYNOPSIS IS NOT A REQUEST FOR COMPETITIVE PROPOSALS. However, interested parties may identify their interest and capability to respond to this notice. A determination by the Government not to compete this proposed contract based upon responses to this notice is solely within the discretion of the Government. All responsible sources may submit a capability statement, proposal, or quotation which shall be considered by the agency. The information received will normally be considered solely for the purposes of determining whether to proceed on a non-competitive basis or to conduct a competitive procurement. Responses to this notice shall contain sufficient information to establish the interested parties' bona-fide capabilities for fulfilling the requirement and include: unit price, list price, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Unique Entity Identifier ID (UEI), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov. All responses must be received by the closing date and time of this announcement and must reference the solicitation number, NIMH-22-005125-NOI. Responses must be submitted electronically to Kyle Miller, Contract Specialist, at kyle.miller2@nih.gov and Christine Frate, Contracting Officer, at christine.frate@nih.gov. U.S. Mail and fax responses will not be accepted.
