NON-COMPETITIVE COMBINED SYNOPSIS / SOLICITATION RFQ- 75N95023Q00188 Title: Single-Nuclei Multiome Analysis of Human Brain Tissue, iPSC-Derived Cells and Human Dorsal Root Ganglion �(i)������� This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (ii)������� The solicitation number is 75N95023Q00188 and the solicitation is issued as a request for quotation (RFQ). ����������� This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures; FAR Subpart 13.5� Simplified Procedures for Certain Commercial Items; and FAR Part 12�Acquisition of Commercial Items, and is expected to exceed the simplified acquisition threshold.� THIS IS A NON-COMPETITIVE (NOTICE OF INTENT) COMBINED SYNOPSIS SOLICITATION TO AWARD A CONTRACT OR PURCHASE ORDER WITHOUT PROVIDING FOR FULL OR OPEN COMPETITION (INCLUDING BRAND-NAME).� The National Institute on Drug Abuse (NIDA), Office of Acquisition (OA), on behalf of the National Center for Advancing Translational Sciences (NCATS), intends to negotiate and award a purchase order without providing for full and open competition (including brand-name) to Active Motif, Inc. for single-nuclei multiome services to be performed by the vendor using 50 samples of human brain tissue, iPSC-derived cells or human dorsal root ganglion. This acquisition is conducted as non-competitive for a commercial item or service and is conducted pursuant to FAR Subpart 13.5�Simplified Procedures for Certain Commercial Items and FAR 13.501, Special Documentation Requirements, in accordance with 41 United States Code (U.S.C.) 1901.� ��Pursuant to FAR Subpart 13.501(a)(1)(iii) the justification (excluding brand-name) will be made available within 14 days after contract award or in the case of unusual and compelling urgency within 30 days after contract award.� (iii)������ The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) Number 2023-02, with effective date 3/16/23. (iv)������ The associated NAICS code 621511 and the small business size standard $41.5 million� This requirement has no set-aside restrictions. (v)������� This requirement is for the following single-nuclei multiome services to be performed by the vendor using 50 samples of human brain tissue, iPSC-derived cells or human dorsal root ganglion. The National Institutes of Health (NIH) is the nation�s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people�s health and save lives. The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), with the mission to catalyze the generation of innovative methods and technologies that will enhance the development, testing and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions. The Stem Cell Translation Laboratory (SCTL) is a state-of-the-art research facility within NCATS� Division of Pre-Clinical Innovation dedicated to addressing the scientific and technological challenges in the iPSC field. The SCTL uses an integrated technological approach to better understand pluripotent stem cell biology and thereby leverage drug discovery and regenerative medicine. The SCTL has acquired human brain tissue, dorsal root ganglion and generated relevant iPSC-derived cell types. The single-cell multiome service is an integral part of the suite of analysis methods that will be carried out to construct a high-resolution �map� of the genomic and epigenomic landscape of neural cell types and human derived tissues. The streamline integration of all these analyses through will allow us to generate foundational datasets that will be shared with the scientific community. Independently and not as an agent of the Government, the Contractor shall furnish all the necessary services, qualified personnel, material, equipment, and facilities, not otherwise provided by the Government as needed to perform the Statement of Work below: The SCTL will obtain human tissues or culture the biological material and harvest the cells. The tissues or cell pellets will be flash frozen on dry ice and will be shipped to the vendor on dry ice. Then, the vendor will extract the nuclei from the tissue or cell pellets, perform quality analysis, construct sequencing libraries, perform sequencing on an Illumina platform, perform data analysis, and biological interpretation. Finally, the vendor will make the data available to the SCTL via FTP or HTML download. Single-cell multiome allow for both transcriptome analysis and genome-wide detection of open chromatin at the single cell level. Understanding both the gene expression profile and the chromatin state at single-cell resolution can help identify how epigenetic changes instruct gene expression in distinct cell populations. This technique can be used to identify cell subpopulations with different transcriptome and epigenetic profiles withing complex samples. The SCTL will provide frozen cell pellets, then the vendor will: For Single-nuclei multiome: Prepare nuclei Tn5 tagmentation of open chromatin regions Nuclei loaded onto 10X Genomics Chromium Controller Barcoding of mRNA and in the same nucleus Library generation Sequencing Bioinformatic analysis The vendor will be responsible for carrying out nuclear extraction, QC of the of the nucleic acid samples, sequencing, and data analysis. The SCTL will derive a total of 50 samples, and ship them to the vendor for analysis. Deliverables: � Raw data (FASTQ) output files. � Aligned data (BAM) files. � Differential methylation analysis tables. � Figures & graphs � Includes coverage depth analysis, heatmaps & pie charts. All reports will be provided by the vendor in electronic form � PDF, MS Excel sheets, MS Word, TXT files, PNG, TIFF figures, or other format based on the type of report. Raw sequencing data will be provided in the form of FASTQ files and will be furnished using as an FTP or HTML server download. (vii)����� The Government anticipates award of a firm fixed-price purchase order for this acquisition, and the anticipated period of performance will commence within 30 days after receipt of order (ARO) and last 4-8 weeks from receipt of the samples intended for sequencing. � (viii)���� The provision at FAR 52.252-1, Solicitation Provisions Incorporated by Reference (Feb 1998), applies to this acquisition. The Government will evaluate offers for award purposes by adding the total price for all options to the total price for the basic requirement. (xiv)���� Responses to this solicitation must include clear and convincing evidence of the offeror�s capability of fulfilling the requirement as it relates to the technical evaluation criteria. The price proposal must include the labor categories, an estimate of the number of hours required for each labor category, fully loaded fixed hourly rate or each labor category, breakdown and rationale for other direct costs or materials, and the total amount. The Unique Entity ID from SAM.gov, the Taxpayer Identification Number (TIN), and the certification of business size must be included in the response. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov. All quotations must be received by 3:00pm Eastern Time on April 7, 2023 and must reference the solicitation number cited above.
