Description
(i)������� This is a combined synopsis/solicitation for commercial items prepared in accordance with the format in Subpart 12.6 as supplemented with additional information included in this notice. This announcement constitutes the only solicitation; proposals are being requested and a written solicitation will not be issued. (ii)������� The solicitation number is 75N95023Q00299 and the solicitation is issued as a request for quotations. This acquisition is for a commercial item or service and is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13�Simplified Acquisition Procedures; FAR Subpart 13.5� Simplified Procedures for Certain Commercial Items; and FAR Part 12�Acquisition of Commercial Items and is expected to exceed the simplified acquisition threshold.� (iii)������ The solicitation document and incorporated provisions and clauses are those in effect through Federal Acquisition Circular (FAC) Number 2023-02, with effective date March 16, 2023. (iv)������ The associated NAICS code 541990 and the small business size standard is $19.5M.�� This requirement is full and open with no set-aside restrictions. (v)������� This requirement is for the following services: DEL screening service to accelerate the discovery of small molecules of biological interest to the program. The DEL screening will be divided into two parts: the first part will involve the assessment of our protein targets� feasibility for the DEL screening; and upon successful reports, the second part will consist of screening the DNA encoded library with the objective of identifying potential lead compounds that can be followed up with medicinal chemistry studies. Scientists and/or technicians with experience in DEL screening shall run the experiment. PART 1: The contractor will run an initial test for a comprehensive review of DEL screening suitability/feasibility. This experiment will produce a fast-turnaround feasibility assessment of protein reagents provided by NCATS to the contractor for validation prior to screening. With this testing we will also have a process to potentially salvage previously failed targets. For each assessment, the contractor will screen a small portion of the target against their DEL library deck, which features several billion compounds. Within the agreed upon period of performance, the contractor will provide the number of prospective high-quality target engagers and assess the target�s output to provide expert recommendations. This will help us decide which targets to prioritize and de-risk our decision making to ensure the best results. To proceed to PART 2, the feasibility result must show targets provided by NCATS are displaying a high number of highly enriched clusters with structure�activity relationship (SAR). These clusters make it possible to quickly navigate and determine the targets suitability to progress to the DEL screen. PART 2: The contractor will run the DEL screening, based on NCATS� guidance and results from PART 1, which is an experiment to test our purified proteins and cell lysate samples against a DNA Encoded Library comprising of billions of compounds. This screening will provide us with a comprehensive report of hits for medicinal chemistry follow up. For each previously assessed protein/cell lysate, the contractor will screen the target against their DEL deck. Within the agreed upon period of performance, the contractor will provide the number of prospective high-quality target engagers and assess the target�s output to provide expert recommendations. The contractor will: Design and synthesize DNA encoded libraries. Preferably this will contain several billion compounds (7 � 8 billion compounds). Run affinity screening of libraries to ensure enrich for selective binders. Characterize selection output using deep sequencing of DNA tags. Identify enriched library compounds through statistical analysis, which will produce the final report NCATS will receive. (vi)������ Background Information and Objective: ����������� The National Center for Advancing Translational Sciences (NCATS) is part of the National Institutes of Health (NIH), with a mission to catalyze the generation of innovative methods and technologies that will enhance the development, testing, and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions. The Early Translational Branch (ETB) is tasked with identifying and developing novel small molecule leads against a wide spectrum of human disease targets, which it accomplishes by screening thousands of small molecules in collaboration with both intra-and extramural researchers. In pursuit of this goal, discovering small molecules that could serve as starting points for therapeutics is of great interest within NCATS. Medicinal chemistry optimization is often an important exercise in our development of small molecules to test the therapeutic hypothesis in disease models. NCATS uses several strategies and modalities to address many aspects of design approaches to discover novel small molecules. One such strategies is the use of DNA-encoded chemical libraries (DELs) which represent a versatile platform for the discovery of small-molecule ligands to protein targets of biological interest. DELs are collections of molecules, individually coupled to distinctive DNA tags serving as amplifiable identification barcodes. Thanks to advances in DNA-compatible reactions, selection methodologies, next-generation sequencing, and data analysis, DEL technology allows the construction and screening of libraries of unprecedented size; this has led to the discovery of highly potent ligands, some of which have progressed to clinical trials. Part of the process is evaluating the targets� feasibility and suitability for a DEL screen. As part of this initial step, NCATS will send 12 purified proteins or cell lysates for testing. Once the feasibility of these samples is confirmed, NCATS will prioritize and select the six best targets to move forward to the DEL screen.� Successful DEL screens will allow NCATS to find more suitable drug leads for our target disease models we are studying. Government Responsibilities: Part 1: NCATS will prepare and send 12 purified proteins or cell lysates samples to the contractor to conduct the study. The contractor will be required to provide a report that includes test results (feasibility assessment) for the 12 purified proteins or cell lysates provided by NCATS. The report shall include the feasibility results in detail. Part 2: Input, feedback on feasibility test. NCATS will select six purified proteins or cell lysates for the full DEL screen. Deliverables: Part 1: Electronic copy of report as indicated in reporting requirements. Part 2: Electronic copy of report as indicated in reporting requirements. Specific Selection/Technical Evaluation Criteria: Offeror must have a Master Service Agreement (MSA) already in effect with NCATS by the time of quote submission. (vii)����� The Government anticipates award of a firm fixed-price contract for this acquisition, and the anticipated period of performance is 1-2 months ARO for Part 1 and 2-4 months from acceptance of Part 1 for the Part 2 portion.
