Description
Statement of Need and Purpose: The NINDS Membrane Transport Biophysics Section (MTBS) requires one (1) INVERTED MANUAL MICROSCOPY SYSTEM to capture high-quality images of live cells and lysosomes while simultaneously conducting patch clamp electrophysiology. This new system will allow for patch-clamp electrophysiology and microscopy techniques to simultaneously detect cellular and pH changes along with ionic current across the membrane, understanding the functions of the lysosome and the role of disease-causing mutations such as in the CLCn7. Background Information and Objective: The National Institute of Neurological Disorders and Stroke (NINDS)-Membrane Biophysics Section (MTBS) studies membrane proteins and transport. We conduct experiments examining the function of lysosomes, from acidification and ion fluxes to cell metabolism and amino acid equilibrium. It is imperative that we have this system to help us better understand the functions of the lysosome and the role of disease-causing mutations such as in the CLCn7 that we are currently studying. This system will allow for simultaneous detection of the pH inside the cell using fluorescence microscopy at the same time that total electric current is being recorded. With a greater understanding of how disease mutations are effecting the lysosomal environment, we hope to be able to help patients to have a better quality of life.
