Description
Specification / Configuration Background Information: A major focus of the Shih lab is to understand the gene regulation of immune cells during neurodegenerative diseases. To achieve a facet of this goal, we need to screen multiple organ systems, including spleen, liver, and brain tissue containing infiltrating peripheral immune cells, and sort them out (isolate) them from surrounding tissue. Justification Rationale for the request: The Shih lab has many transgenic mice lines in buildings 6B and 35A. Currently, we do not have a dedicated cell sorter, and we must rely on sorters from various core institutes at the NIH to perform these sorts. We will routinely need the use of a cell sorter to complete our projects, and our workload has steadily increased, meaning increased sort times at the IC cores. The increased usage has made it impractical not to have a dedicated sorter in the lab. The Sony SH800 Cell Sorter uses a disposable microfluidic nozzle for size-selected sorting while decreasing sample contamination. Additionally, the SH800 has four lasers, which allows for a greater range of fluorescent sorting. Another added benefit of the SH800 over other sorters of similar functions is the fully integrated platform that makes this sorter user-friendly with minimal training. The SH800 has fully automated calibration, alignment and focusing, monitoring, and sorting. This integrated automation allows for minimally trained users to operate the sorter compared to other sorters, which need dedicated personnel to operate. Moreover, the SH800 Cell Sorter is small and can be placed on any lab benchtop versus conventional sorters that require an entire room for their use; thus, no additional lab space or alterations will be needed.
