Army Medical Research and Materiel Command is currently seeking basic research proposals to develop an immunoassay for quantitating thiodiglycol (2,2'-thiodiethanol) in urine and other biological fluids. The immunoassay should demonstrate the specificity and sensitivity necessary to be used as a screen for thiodiglycol in urine samples and biological fluids. Also, the immunoassay method should be simple and portable for use in the field. The offerer is expected to have expertise in monoclonal antibody technology. In addition, the offerer's facility should be currently fully operational for such work. While the offerer chosen for award of any contract is not limited to use Government supplied haptens to aid in the development of a monoclonal antibody for thiodiglycol, the offerer may obtain from the Government four haptens to use in preparing test antigens and immunizing antigens. Available Government supplied haptens are: (a) Set 1. 4-(2-hydroxyethyl) benzoic acid for the preparation of an immunizing antigen and 8-hydroxyoctanoic acid for the preparation of the test antigen, and (b) Set 2. 4-(2-hydroxyethylthio) benzoic acid for the preparation of an immunizing antigen and 6-(2-hydroxyethylthio) hexanoic acid for the preparation of the test antigen. Following both contractor and Army characterization of the monoclonal antibodies produced by the offerer, the offerer will produce 500 milligrams of the monoclonal antibody selected by the Army. All cell lines developed during the investigation will become the property of the Army following completion of the project. Proposers are encouraged to survey the open literature on this topic prior to submitting their research ideas in a preproposal for Army review. This research will support a program on medical countermeasures against vesicating chemical warfare agents.
