The National Institutes of Health (NIH), National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) intends to purchase The PHOTOMETRIC DIGITAL CHARGED COUPLED DEVICE COOLED CATHODE DETECTOR (CCD) CAMERA, to work together with our existing microscope imaging system. This will provide a sensitive method for detecting nuclear import of viral DNA. Our current camera is not sensitive enough to detect single photons emitted by bioluminescent reactions. We will alter the gene encoding for the firefly luciferase enzyme to contain a nuclear localization signal. Upon expression of the gene in cultured cells, luciferase will accumulate within the nucleus. Then, luciferin can be conjugated to proteins or with biotinylated DNA and can serve as a reporter. The movement of viral DNA across the nuclear membrane will be detected by the light emission along the path. Anti-viral agents can be tested to inhibit nuclear transport of viral DNA. This camera can also be used to detect steroid receptor movement across the nuclear membrane, and rapid changes in intracellular calcium levels, known to regulate nuclear transport. Universal Imaging will provide free software, installation and training for this upgrade of our existing Universal Imaging microscope imaging system. The Justification For Other Than Full And Open Competition is that this camera has to work with two (2) microscopes: a Zeiss Axiovert 100 & with Zeiss Photomicroscope 3. The ultra high sensitivity and the low noise level of the camera is essential for bioluminescence measurement of biologic events emitting single photons. The intended source Universal Imaging is the manufacture of the Metamorph software that is customized for The PHOTOMETRIC Digital CHARGED COUPLED DEVICE COOLED CATHODE DETECTOR (CCD) CAMERA SYSTEM. Universal Imaging will integrate The PHOTOMETRIC DIGITAL Charged COUPLED DEVICE COOLED CATHODE DETECTOR (CCD) CAMERA SYSTEM into a computer control system (Universal Imaging Metamorph and MACINTOSH IP Lab) driving Ludle filter wheel, X,Y,Z stage controllers currently used with these microscopes. Any adaptor to the microscope should be included. Maintenance agreements should be available for an extended period of time. All responsible sources may submit a quotation, which if timely received, shall be considered by the agency. (See Note 22). It is anticipated that Request For Quotation (RFQ) Number NIDDK-SSAS-95-111 will be available fifteen (15) days after the publication date of this synopsis. Receipt of quotations will be due ten (10) days after the release of the RFQ. The anticipated award date is September 11, 1995.
