National Institues of Diabetes and Digestive and Kidney Diseases has
a requirement for the development of a Mouse Knock-In for fragile X
syndrome on a competitive basis. The fragile X mental retardation from
in humans is due to an expansion of CGG trinucleotide repeats within
the FMR1 gene which results in the absence of the FMR1 protein (FMRP).
We would like to develop a mouse model for the triplet expansion
defect by inserting a large number of CGG repeats into the CGG tandem
array in the 5 UTR of the mouse FMR1 gene. The following milestones
must be achieved: restriction mapping and exonic sequencing of the
mouse FMR1 genomic clone, engineering of the targeting vector. We will
provide a fragment of multiple (>100) CGG repeats that will need to be
put into the targeting vector, Transfection of the targeting vector
into ES cells and selection of at least 150 ES clones, Screening for
homologous recombination by southern analysis of selecting ES clones.
If no recombination then steps 3 and 4 will be repeated a second time
at no charge to us, Expansion of recombinant clones, and Production of
chimeric animals (at least 3), which will all be shipped to us after
having been MAP tested for pathogens. All responsible sources may
submit a quotation, which if timely received, shall be in within the
Simipified Acquisition Thereshold $ 100,000. It is anticapted that
Request for Quotation (RFQ) NIH-NIDDK-97-305 will be available fifteen
(15) days after the publication date of this synopsis receipt of
quotations will be due ten (10) days after the release of the RFQ. The
anticapted award date is September 26, 1997. Requests for copies of
