66 -- INSTRUMENTS & LABORATORY EQUIPMENT SOL NCI-RFQ-70200 DUE 092497 POC Cynthia Brown, Purchasing Agent, 301-402-4509, Patricia Haun, Contracting Officer Under the authority of 41.U.S.C. 253(c)(1), FAR 6.302-1, the National Institutes of Health, National Cancer Institutes intends to procure from Pharmacia Biotech Inc., P.O. Box 18191, Newark, N.J. 07191, one(1) AKTA Explorer-10 System/Liquid Chromatography System and Accessories. The Liquid Chromatography System is required to: (1) Identify tumor-associated antigens recognized by T helper cells by purifying individual immunostimulatory proteins from complex lysates of whole tumor cells; (2) Purifying known tumor antigens recognized by T cells, as expressed in recombinant bacterial or eukaryotic systems, for study in animal models of protein vaccination, for use as in vitro immune stimulants in human or murine systems, or as reagents for raising antibodies reactive with these proteins in rodents; (3) Purifying individual immunostimulatory tumor-associated peptides following synthesis, for use in animal models of peptide vaccination or for use in raising reactive T cells by in vitro repetitive stimulation. The flow range of 0.001 to 10ml/min, and the operating pressure range of 0-25 Mpa are specifically designed to provide analytical scale HPLC as well as preparative scale FPLC chromatography functions, thus eliminating the need to purchase two separate chromatography units to serve the specific requirements. The AKTA Explorer-10 System will give investigators the ability to purify biomolecules from complex starting materials derived from cell extracts, tissue homogenates and recombinant protein expression systems as well as to purify synthetic peptides, polypeptides and oligonucleotides through separations based on a variety of sample characteristics such as molecular weight, net surface charge and hydrophobicity, while retaining biological activity. The system also provides the unique ability to reverse the direction of solvent flow in this system so that the desired chromatography column may be selected and installed without removing tubing and fittings by which the column is connected to the flow path. Removal is time consuming and also risks introducing air bubbles into the system. The AKTA Explorer-10 System is compatible with a variety of chromatographic columns and media already obtained from Pharmacia, such as RP-HPLC columns (Sephasil Peptide C-18, Sephasil Protein C-4, Thompson C-8), ion exchange columns (Resource Q, Hi Trap Q, Hi Trap SP, TosoHaas DEAE-5PVW), and gel filtration columns (TosoHaas G 3000 SW, 21.5 mm and 7.8mm). This notice of intent is not a request for competitive proposals as no solicitation will be issued. However, vendors who believe they can meet the Government's requirements, including compatability with existing chromatographic columns and media may submit their qualifications. The submission must be in writing and must contain sufficient detail so that a determinatin can be made regarding meeting the Government's requirements.
