giffordf@ars.usda.gov. The USDA-ARS in Albany, Ca. has a requirement
for a FPLC system which can perform as follows: The system must perform
separations based on a variety of sample characteristics molecular
weight, net surface charge, hydrophobicity, while retaining biological
activity. It should have large open column 0.05-20mls/min flow rate,
two pump gradient system, control of up to nine motorized valves (one
automatic three position valve must be included) for control of column
selection, buffer selection, flow reversal and multi-dimensional
chromatography. The entire wetted flow path must be inert to halides.
The system must automatically and continuously record for the complete
run the flow rate, auto scaled UV signals, actual and programmed
conductivity of the eluent, eluent ph, and system pressure, accumulated
time of run, all valve positions and changes in position, activity of
fraction collector. Valves must be driven electronically, with no
external air pressure required. The column equilibration feed-back must
be monitored. The controller must allow programs to be time based,
volume based & column volume based. The system must be a two pump
gradient system, with dynamic mixing. A minimum 100 position fraction
collector controllable from the system software must be included.
Control of chart recorder, fraction collector, gradient mixer, air
sensors, and three pumps from the system software. Equipment must be
operational in a cold room at 4C. Post run chromatographic analysis by
software. Documentation of runs should be automatic and complete. The
data must be in a form for export to a spreadsheet. Column definable
pressure limit which stops pump & sets off an alarm. The equipment
listed below will meet our requirement but alternate source will
certainly be considered. Installation and training must be provided by
the vendor at no additional cost. The system quoted should include
spare parts likely to be needed in the first 12 months of operation. 01
