A -- SERVICES SOL RFQ80245(NR) DUE 092598 POC Marsha Gorham, Purchasing Agent, (301)402-4509 or Todd Cole, Contracting Officer The Environmental Epidemiology Branch (EEB), Division of Cancer Epidemiology and Genetics (DCEG), National Cancer Institute(NCI) plans to procure the services of Johns Hopkins University, 600 North Wolfe Street/CMSC 604, Baltimore, MD 21287-3654. The purpose of this requirement is to provide support to extract DNA and perform genetic assays in blood spot samples from 245 girls 16-18 years of age participating in the Dietary Intervention Study in Children. The DISC study is a National Heart Lung and Blood Institute (NHLBI) sponsored, multicenter clinical trial that evaluates the efficacy of a fat-modified diet to lower LDL-cholesterol in children and the adequacy of diet to promote growth and development. Based on work conducted under the DISC study, the study herein will evaluate the influence of genetic polymorphisms in steroid synthesis and metabolizing enzymes (CYP11a, CYP17, COMT, and CYP1B1) on serum levels of sex hormones. Genetic assays will be performed using blood spots from the 245 participating girls that were collected from Johns Hopkins University as part of a Gene/Diet Acillary Study. Serum levels of sex-hormones, which correspond to the subject blood spot samples, have already been determined as part of the DISC Hormone Ancillary Study. Additionally, the blood spot samples are the property of Johns Hopkins University. It should also be noted that Johns Hopkins University has extensive experience in extracting DNA from blood spots and performing assays to determine genetic polymorphisms from extracted DNA. Since the existing data from the DISC study provides a unique opportunity to study the genetic control of serum hormone levels and the blood spot samples have already been collected and serum hormone levels established, Johns Hopkins University is the only source known to NCI that can meet all of the above requirements. If any interested party believes it can perform the above requirement it may submit a statement of capabilities. The statement of capabilities and any other information furnished must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can perform this requirement.
