Devices and Radiological Health [CDRH], intends to award a purchase
order to Pathology Associates International based on the Scope of Work
for technical services to complete the following tasks: 1] Perform
gross and histologic examination of fetal/embryonic tissue to assay
segmental and other defects, 2] Analyze embryonic tissues for
qualitative and quantitative changes in genetic expression and identify
unique proteins that are expressed in response to physical and chemical
exposure using gel electrophoresis technicques,
immunochemical/immunohistochemical methodologies, 3] Perform assays to
identify alterations in the proliferation cells with the presomitic
mesoderm and other issues of the developing embryo following in vivo
and in vitro hypertermia treatment. These assays will incorporate
immunocytochemical techniques such as detection of prolifering cell
nuclear antigen and/or in situ hybridization for histone mRNA, 4]
Analyze embryonic tissues for the specific DNA endonuclease activity
that is accelerated during programmed cell death using some of the
following techniques: endonuclease activation, gel electrophoresis for
DNA fragmentation, in situ hybridization and quantitative RT-PCR, and
5] Analyze embryonic tissues for identification of unique gene
expression following hyperthermia using termo-stable DNA polymerase and
specific primers for a specific sequellnce or gene of interest [heat
shock genes/proteins; homeobox genes/proteins, etc.]. Period of
