Specifications for Gel and Blot Analyses 1. One system must be capable
of scanning, via software control, storage phosphor screens
(autoradiography), red and blue excited fluorescent of DNA and protein
gels, and chemifluorescent DNA/RNA and protein blots. 2.The selection
of the appropriate optical components must be automatic when one of
the three scanning modes is selected. System must use storage phospor
screen technology, not flow through proportional counting. 3. Storage
phosphor screens must be available unmounted in standard film cassettes
and mounted on aluminum backing. 4. System must be compatible with
existing Phosphorlmager storage phosphor screens and cassettes used for
Molecular Dynamics 400E Phosphorimager. 5. System must be able to
utilize 35x43 cm and 20x25 cm format mounted and unmounted storage
screens, as well as gels and blots. 6. Scanning resolution must range
from 200 to 50microns in software selectable units, with a beam
diameter as small as 50 microns. 7. Uniformity must be plus or minus 5%
relative standard deviation, and pixel accuracy plus or minus 0.15%.
8.Linearity must be less than 7.5% relative standard deviation for the
entire dynamic range. 9. Linear dynamic range must be 5 orders of
magnitude. 10. Light measurement for storage phosphor, fluorescence,
and chemifluorescence must be collected and converted to electrical
signal by a photomultiplier detector. 11. Light source is desireable to
be a laser diode at 635 nm and an LED at 450 nm, with average lifetime
of at least 50 thousand hours. 12. Detection thresholds: lower limit
for one hour exposure must be less than 2 dpm/mm squared for
Carbon-14(for 200 and 100 micron pixel size), with lower limit for
detection of Phosphorous-32 usually at least five times lower. 13. Two
excitation sources in one instrument: a. 635 nm excitation source for
scanning phosphor screens and red-excited florescent samples. b. 450
nm excitation source for blue excited fluorescence and chemifluorescent
samples. 14.Would like one software program to control the instrument,
scan the sample,acquire the image, and analyze the data from all three
scanning modes. 15. Software to perform operations identified in
requirement 14, must be ab;e tp run on Macintosh PowerPC computers with
G3 and 604e 200 MHz CPUs. 16/Updates to software must be provided at no
additional charge during the warranty period. Warranty must be covered
by at least one year. 17 System must include an image-eraser box for
all size storage screens used by the system. 18 System and
image-erasure box must operate on 50/60 Hz and 115/230V (switchable)
with power draw of no more than 500 watts for the system and 150 watts
for the image erasure. 19. System and image-erasure dimensions in
inches should not exceed: height width depth system 20 35 31
