A -- EVALUATION OF THE EFFECT OF VACCINATION WITH ESCHERICHIA COLI 0157 LOPOPOLYSACCHARIDE TO PREVENT COLONIZATION OF CALVES WITH THIS HUMAN PATHOGEN. SOL NICHD-DIR-99-11 POC Mrs. Mya N. The National Institutes of Child Health and Human Development, National Institutes of Health plans to award a contract to the University of Idaho, using other than full and open competition to evaluate the safety and efficacy of E. coli 0157 for prevention of colonization of young cows (calves) with this human pathogen. A Phase I study of E. coli 0157 conjugate vaccine in healthy adults, demonstrating safety and immunogenicity of our investigational vaccine has been published. A manuscript describing the immunogenicity of our 0157 conjugates in young cows (calves) compared to the lipopolysaccharide (LPS) from this pathogen is in preparation. The determination to use other than full and open competition was made in accordance with USC 253 (c)(1) and is based upon the following factors: 1) The proposed Principal Investigator has maintained a broad and continual interest in E.coli in cows. She has been trained in the very best laboratory concerned with basic aspects of bacterial toxins of enteric pathogens including the Shigella-like toxin of E. coli 0157. She has studied the serology of animals fed E.coli 0157 and the effect of this intervention upon colonization with this pathogen. 2) The proposed contractor is the only organization that has the facilities and trained personnel to maintain an ongoing and successful program in studying the induction, detection, and effects of various interventions upon colonization of cattle and other domestic animals with E. coli 0157 in order to achieve 0157-free herds. The purpose of this synopsis is to determine whether there are other sources with the above capabilities to perform the work described above.
