The U.S. Environmental Protection Agency (EPA) intends to negotiate sole source procurement with Stratagene Corporation for the acquisition of a Purification of RNA from tissue/cells and a Lambda Zap II Vector custom library. The custom Zap II library also includes amplification of one million primary recombinant clones, host strains for growth and screening, helper phage for excisions and rescues, and protocols for screening and DNA manipulation. The contractor shall provide the following: (1) purify cDNA from the tissue provided and furnish convincing evidence of the integrity of the isolated RNA by supplying photographic documentation of the integrity of RNA after gel electrophoresis; (2)generate cDNA from the RNA and clone the cDNA into the appropriate vector to generate libraries; (3)provide amplified and unamplified versions of each library;(4) provide information as to the average size of the insert in each library and shall investigate the length of at least ten clones to obtain this information and (5)deliver amplified and unamplified libraries, all requested supporting documentation, and the remaining starting material as well as any isolated RNA which was not used in library construction. EPA believes Stratagene Corporation is the only source of custom library construction which utilizes the lambda zap phage backbone. The use of lambda zap backbone is proprietary and provided only by Stratagene. Any firm which feels it is capable of satisfying the requirements stated above should fully explain their capabilities in writing within 15 days of the date of this announcement.
