Agent 301-594-7731 The National Institutes of Health (NIH) National
Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) has a
requirement to purchase the services of a vendor for the Production of
Knockout Transgenic mice. This requirement will be used to produce a
cre/lox dependent knockout mouse removing the O-linked GlcNAc
Transferase locus. This is part of a long-term effort to characterize
this locus and define its role in type II diabetes and the functioning
of the islet cells of the pancreas. The government laboratory will
supply the BAC (Bacterial Artificial Chromosome) clone containing the
OGT gene and provide sequence and positional information regarding the
locus of interest. This will included confidential sequence
information and a general strategy for the production of the targeting
vector. The Government will supply any information required for
production of probes and other items required for the production. The
vendor will be required to 1) Generally Map the OGT gene in sufficient
detail to make a targeting vector 2) Prepare a targeting vector
(cre/lox) which will allow gene disruption 3) Introduce the vector into
recipient Embryonic Stem cells by genetic selection 4) Inject the stem
cells into psuedo pregnant females for production of chimeras 5) Breed
to strain insuring germline transmission of the knockout allele 6)
Breed knockout strain to homogygosity 7) Provide the government with 5
breeding pairs of mice carrying the mutant allele for subsequent
breeding to a targeted cre expressing mouse strain. 8) A laboratory
facility that is AALAC approved or has their approved pathogen-free
animal facility. It is anticipated that these steps should be completed
by the vendor within 6-8 months of initial contact with the government
laboratory. The Standard Industrial Classification Code is 3011 and
the business size standard is 500. All responsible sources may submit
a quotation, which if timely received, shall be within the Simplified
Acquisition Threshold for Purchases up $100,000. It is the intent of
the National Institute of Diabetes and Digestive and Kidney Disease to
purchase the required laboratory services specified in the Request for
Quote (RFQ). Vendors capable of furnishing the government this service
should request a copy of the request for Quote form from the above
contact. It is anticipated that Request for Quotation (RFQ)
NIH-NIDDK-99-480 will be available fifteen (15) days after the
publication date of this synopsis. Receipt of quotations will be due
ten (10) days after the publication date of this synopsis. The
anticipated award date is September 27, 1999. Request for copies must
