66 -- DYNAPRO-MS800 W/ ATTENUATING LASER TEMPERATURE CONTROLLER SOL NCI-RFQ-90237-NR DUE 092419 POC Marsha Gorham, Purchasing Agent & Todd Cole, Contracting Officer (301)402-4509 The National Cancer Institute (NCI), Division of Basic Sciences (DBS), Laboratory of Cell Biology (LCB) plans on purchasing a DynaPro-MS800 with attenuating Laser Temperature Controller (with Windows 98 based PC) from Protein Solutions, Inc., 1224 West Main Street, Suite 777, Charlottesville, VA 22903. The Laboratory of Cell Biology is dedicated to understanding the molecular machanisms by which ATPases promote biologically important energy-requiring processes involved in ligand transport, protein translocation, and protein unfolding. In performing this research, the lab conducts solution studies of purified proteins and protein complexes. The aforementioned instrument is required since light scattering measurements will be made on purified proteins at high concentrations as a check on quality for crystallization purposes. Light scattering will be used for short term dynamic measurements of proteins. This instrument will greatly expand our ability to understand the interactions between the protein components that compose the ATP-dependent proteases studied in the laboratory. Of particular significance will be measurements that provide information about the rates at which protein conformations and oligomeric states change during catalysis. The required instrument shall allow mesurements, data collection, and analysis of solution properties of proteins. In addition, the instrument shall be capable of being used in conjunction with existing fluorescence and circular dichroism instrumentation. It shall be capable of providing details about oligomeric structure, assembly and disassembly, and conformational changes in the components in response to ligand binding and their impact on the enzymatic properties of the structures. Specifically, this light scattering instrument shall have the following features/characteristics: 1) Shall provide, accurate reproducible measurements without damage to the proteins; 2) Shall be capable of detecting dynamic changes in the protein over a broad range of temperatures; 3) There shall be precise temperature control of the sample cuvette over a range of temperatures from 5 degrees Celsius to 65 degrees Celsius; 4) Shall be able to detect at high sensitivity solutes in the size range of 25,000 Da to greater than 1 x 10 to the sixth Da; 5) Shall have sensitivity over a wide range of solute concentrations from 20 micrograms per ml to greater than 10 milligrams per ml; 6) Shall enable very small sample volumes to be measured; 7) Shall have a design that allows facile addition and mixing of samples in the cuvette; 8) Shall have the capability to perform time based measurements of light scattering; and 9) Shall have software support to allow accurate and easy data analysis and display. Protein Solutions, Inc. is the only source known to the NCI Researcher that can provide light scattering instrumentation, which allows measurements, data collection, and analysis of solution properties of proteins, and that can meet the specific requirements above, especially with regard to measuring particles in the size range noted. This notice of intent is not a request for competitive quotation. However, if any interested party believes it can meet the above upgrade requirement, it may submit a statement of capabilities. The capability statement and any other furnished information must be in writing and must contain material in sufficient detail to allow NCI to determine if the party can fully meet the requirements.
