National Cancer Institute (NCI), Division of Clinical Sciences (DCS),
Medicine Branch (MB) plans on procuring a four color flow cytometer,
the FACSCalibur, from Becton Dickinson Immunocytometry Systems, 2350
Qume Drive, San Jose, CA 95131. The Medicine Branch has been involved
in the development of patient-specific idiotype vaccines for human
follicular lymphoma and multiple myeloma patients. The patients receive
the vaccine in the Medicine Branch . The addition of a four color flow
cytometer will enable the Medicine Branch to perform additional
investigations, including: 1) Analysis of intracellular cytokine
profiles in fresh PBMC prepared from patients under vaccine therapy
during scheduled visits and in the tumor-specific T cells generated in
vitro from these patients; 2) Phenotype analysis of biopsy tumor
specimens of interest; 3) Phenotype analysis of ID-specific T cells
generated from lymphoma and myeloma patients; 4) Mapping of antigenic
epitope(s) specific for individual patient tumors; 5) Characterization
of dendritic cells generated from patients/donors under the bone
marrow transplantation protocol; and 6) Sorting of CD8+ T cells from
fresh PBMC and tumor antigen-simulated primary PBMC cultures to
establish antigen-specific CTL. The Medicine branch requires a four
color flow cytometer for the above studies. Specifically, the cytometer
shall have the following characteristics and features: 1) Shall enable
multiparameter flow cytometry analysis of different cell populations;
2) Shall have an Argon Ion Laser and a compatible diode laser to
provide measurement of four colors (four fluorescent channels); 3)
Shall allow the four color measurement in addition to the forward and
side scatter parameters; 4) Shall be a benchtop, modular design but
adequately support dual laser operation; 5) Shall allow for maximum
fluorochrome and antibody choices by providing five or more
photomultiplier tubes; 6) The dual laser beams shall be alignment free
with spatial and temporal separation of the beams; 7) Instrument shall
insure automatic electronic separation of spectra overlap and provide
accurate measurements of fluorescence; 8) The instrument flow cell
shall be at least 480 by 400 microns; and 9) Shall have a Macintosh
Power PC, monitor, and a laser printer. Becton Dickinson is the only
source known to the NCI Researcher that can meet the above
specifications. This notice of intent is not a request for competitive
quotation. However, if any interested party believes it can meet the
above requirement, it may submit a statement of capabilities. The
capability statement and any other furnished information must be in
writing and must contain material in sufficient detail to allow NCI to
determine if the party can fully meet the requirements. Capability
statements must be received in the contracting office by 3:00 p.m., EST
